Duloxetine, a Balanced Serotonin-Norepinephrine Reuptake Inhibitor, Improves Painful Chemotherapy-Induced Peripheral Neuropathy by Inhibiting Activation of p38 MAPK and NF- κB
Conclusion Our results suggested that duloxetine may be an effective treatment for OXA- and PTX-induced peripheral neuropathy. Furthermore, we showed that duloxetine did not reduce the chemotherapeutic efficacy of PTX or OXA. To our knowledge, this is the first report demonstrating the neuroprotective properties of duloxetine both in vitro and in vivo. In addition, we showed that duloxetine may act by blocking activation of the MAPK signaling pathways, thus preventing NF-κB activation and translocation into the nucleus. NGF may have facilitated duloxetine-induced inhibition of nerve degeneration. Our findings sugge...

Structure –Function and Therapeutic Potential of Spider Venom-Derived Cysteine Knot Peptides Targeting Sodium Channels
This study confirmed previous observations that HwTx-IV binds to the voltage-sensor of DII site 4 (Xiao et al., 2008a, 2010) and that ProTx-II binds to the voltage-sensors of DII and DIII sites 4 and 3 (Bosmans et al., 2008; Xiao et al., 2010). The cryo-EM structure of a chimeric hNaV1.7-NaVAb channel bound to ProTx-II was also described recently (Figure 1C) (Xu et al., 2019), which confirmed ProTx-II binds to the voltage-sensor of DII site 4 of the NaV1.7 channel (Xiao et al., 2010). Although the determination of the three-dimensional structure of spider ICK peptides using X-ray or NMR are essential for the understanding ...

75 year old dialysis patient with nausea, vomiting and lightheadedness
Written by Jesse McLaren A 75 year-old patient with diabetes and end stage renal disease was sent to the ED after dialysis for three days of nausea, vomiting, loose stool, lightheadedness and fatigue. RR18 sat 99% HR 90 BP 90/60, afebrile. Below is the 15 lead ECG. What do you think? There ’s normal sinus rhythm, normal conduction, normal axis, normal R wave progression and normal voltages. There’s subtle inferior ST elevation with straightening of the ST segment, reciprocal ST depression and T wave inversion in aVL, and ST depression in V2. This is diagnostic of infero-posterior OMI, but it is falsely n...

Mycolactone as Analgesic: Subcutaneous Bioavailability Parameters
In conclusion, there is no proof yet that mycolactone diffuses in the blood in the context of M. ulcerans infection. Discussion Pain represents currently the most common symptom for which medical attention is sought by patients (Melnikova, 2010). Available treatments have limited effectiveness and significant side-effects (Borsook et al., 2014; Stern and Roberts, 2016). In addition, analgesia duration is often low. Handling of pain remains though a major societal and humanitarian challenge. Mycolactone is a toxin produced by M. ulcerans and responsible for the ulcerative skin lesions observed in patients suffering from ...

Use Profile of Magnesium Sulfate in Anesthesia in Brazil
Conclusions: Magnesium sulfate is an important adjuvant drug in the practice of anesthesia, with several clinical effects and a low incidence of adverse events when used at recommended doses. Introduction Magnesium is the fourth most common ion in the body, and it participates in several cellular processes, including protein synthesis, neuromuscular function and stability of nucleic acid, as well as regulating other electrolytes such as calcium and sodium. Magnesium acts as a cofactor in protein synthesis, neuromuscular function and stability and the function of nucleic acids. It is a component of adenosine 5-triph...

Surprising Research on Cannabis
Much of what we think we know about cannabis may soon change as a result of new research that uncovers some surprising facts. Indeed, the topic, which can be emotionally charged, is the focus of intense scientific study. Is cannabis good for you? Is it addictive? What long-term harms can use cause? The answers to these questions are multi-layered and not always clear-cut, which is why cannabis research continues with even more urgency. FACTS ON CANNABIS ADDICTION AND DEPENDENCE Current estimates are that one in 10 cannabis users will develop cannabis addiction or dependence. The potency of the delta-9-tetrahydrocannabinol ...
