Single-nucleus transcriptomic mapping uncovers targets for traumatic brain injury
In this study, we meticulously mapped the transcriptome atlas of murine SVZ and its responses to mTBI at the single-cell level. We observed cell-specific gene expression changes following mTBI and unveiled diverse cell-to-cell interaction networks that influence a wide array of cellular processes. Moreover, we reported novel neurogenesis lineage trajectories and related key transcription factors, which we subsequently validated through loss-of-function experiments. Specifically, we validated the role of Tcf7l1, a cell cycle gene regulator, in promoting neural stem cell differentiation towards the neuronal lineage after mTB...

The Warburg effect on radioresistance: Survival beyond growth
Biochim Biophys Acta Rev Cancer. 2023 Sep 17:188988. doi: 10.1016/j.bbcan.2023.188988. Online ahead of print.ABSTRACTThe Warburg effect is a phenomenon in which cancer cells rely primarily on glycolysis rather than oxidative phosphorylation, even in the presence of oxygen. Although evidence of its involvement in cell proliferation has been discovered, the advantages of the Warburg effect in cancer cell survival under treatment have not been fully elucidated. In recent years, the metabolic characteristics of radioresistant cancer cells have been evaluated, enabling an extension of the original concept of the Warburg effect....

The Role of Mesenchymal Stem/Stromal Cells Secretome in Macrophage Polarization: Perspectives on Treating Inflammatory Diseases
Curr Stem Cell Res Ther. 2023 Sep 4. doi: 10.2174/1574888X18666230811093101. Online ahead of print.ABSTRACTMesenchymal stem/stromal cells (MSCs) have exhibited potential for treating multiple inflammation- related diseases (IRDs) due to their easy acquisition, unique immunomodulatory and tissue repair properties, and immune-privileged characteristics. It is worth mentioning that MSCs release a wide array of soluble bioactive components in the secretome that modulate host innate and adaptive immune responses and promote the resolution of inflammation. As the first line of defense, macrophages exist throughout the entire inf...

The repair and regeneration mechanism of platelet-rich fibrin-promoting tissue after alveolar bone defect through the notch pathway
Cell Mol Biol (Noisy-le-grand). 2023 Jul 31;69(7):85-90. doi: 10.14715/cmb/2023.69.7.14.ABSTRACTThis work investigated the effects of platelet-rich fibrin (PRF) combined with bone mesenchymal stem cells (BMSCs) on the repair of alveolar bone defect (ABD) and the related mechanism of the Notch1/Wnt3a signaling pathway. 28 healthy male New Zealand white rabbits were selected to prepare the BMSCs and PRF. Rabbits were rolled into a combination group (implanted with PRF + BMSCs for treatment), a PRF group (treated with PRF) a BMSC group (BMSCs for treatment), and a control group (Ctrl group, no material implantation), with 7 r...

Comparative efficacy of endogenous stem cells recruiting hydrogels and stem cell-loaded hydrogels in knee cartilage regeneration: a meta-analysis
Conclusion The current study indicates that both endogenous stem cell recruiting hydrogels and stem cell loaded hydrogels can effectively promote knee joint cartilage regeneration in animal trials.PMID:37711133 | DOI:10.2174/1574888X19666230914123443

Allele-specific expression reveals genetic drivers of tissue regeneration in mice
Cell Stem Cell. 2023 Sep 13:S1934-5909(23)00293-X. doi: 10.1016/j.stem.2023.08.010. Online ahead of print.ABSTRACTIn adult mammals, skin wounds typically heal by scarring rather than through regeneration. In contrast, "super-healer" Murphy Roths Large (MRL) mice have the unusual ability to regenerate ear punch wounds; however, the molecular basis for this regeneration remains elusive. Here, in hybrid crosses between MRL and non-regenerating mice, we used allele-specific gene expression to identify cis-regulatory variation associated with ear regeneration. Analyzing three major cell populations (immune, fibroblast, and endo...
