The two-step approach to allogeneic hematopoietic stem cell transplantation
Front Immunol. 2023 Sep 1;14:1237782. doi: 10.3389/fimmu.2023.1237782. eCollection 2023.ABSTRACTAllogeneic hematopoietic stem cell transplantation (HSCT) provides the only potentially curative option for multiple hematological conditions. However, allogeneic HSCT outcomes rely on an optimal balance of effective immune recovery, minimal graft-versus-host disease (GVHD), and lasting control of disease. The quest to attain this balance has proven challenging over the past few decades. The two-step approach to HSCT was conceptualized and pioneered at Thomas Jefferson University in 2005 and remains the main platform for allogra...

Advances in acute myeloid leukemia differentiation therapy: A critical review
Biochem Pharmacol. 2023 Jul 26:115709. doi: 10.1016/j.bcp.2023.115709. Online ahead of print.ABSTRACTAcute myeloid leukemia (AML) is characterized by impaired differentiation and indefinite proliferation of abnormal myeloid progenitors. Although differentiating agents were deemed to revolutionize AML therapy, most treated non-APL AML patients are refractory or relapse. According to cancer stem cell model, leukemia-initiating cells are the root cause of relapse given their unidirectional potential to generate differentiated AML blasts. Nonetheless, accumulating evidences emphasize the de-differentiation plasticity and leuke...

Why and How Should Ethiopia Establish a Stem Cell Transplant Service? A Review Article
Biologics. 2023 Mar 20;17:33-40. doi: 10.2147/BTT.S401289. eCollection 2023.ABSTRACTEthiopia is attempting to reduce cancer-related morbidity and mortality through a strategic national cancer control plan but according to Globocan 2020, hematologic malignancies particularly leukemia and non-Hodgkin's lymphoma rank among the top five leading causes of new cancer incidence and cause of death among all age groups in both sexes. Hematopoietic stem-cell transplantation (HSCT) is an advanced treatment modality that makes the only effective treatment for cancer and non-cancer-related hematologic diseases unresponsive to conventio...

Lessons learned: a look back at the performance of nine COVID-19 serologic assays and their proposed utility
CONCLUSIONS: Our data suggests against the use of anti-SARS-CoV-2 assays to aid in acute diagnosis. RN TOT and RS TOT can readily identify past-resolved infection and vaccine response in the absence of native infection. We provide an estimate of expected antibody response in healthy VD over the time course of vaccination for which to compare antibody responses in immunosuppressed patients.PMID:36878344 | PMC:PMC9985916 | DOI:10.1016/j.clinbiochem.2023.03.003

A Short History of the Consideration of Sex Differences in Biomedical Research - Lessons for the < em > In Vitro < /em > Community from Animal Models and Human Clinical Trials
Altern Lab Anim. 2023 Feb 7:2611929231156720. doi: 10.1177/02611929231156720. Online ahead of print.ABSTRACTIn recent decades, it has become clear that in many fields - such as drug development, particularly with regard to drug dosage and specific disease treatment - the sex of a patient must be taken into consideration, in view of the fact that male and female physiology and pathophysiology show many differences of practical concern. While, in the last decade or so, considerable efforts have been undertaken to consider the sex of the animals during the planning of experiments, this topic has just started to be acknowledge...

Characterization of cardiac metabolism in iPSC-derived cardiomyocytes: lessons from maturation and disease modeling
CONCLUSIONS: Maturation strategies improve the metabolic characteristics of iPSC-CMs, but not to the level of the adult heart. Therefore, a combination of maturation strategies might prove to be more effective. Due to early onset hypertrophy, HCM iPSC-CMs may be less suitable to detect early disease modifiers in HCM and might prove more useful to examine the effects of gene editing and new drugs in advanced disease stages. With this review, we provide an overview of the assays used for characterization of cardiac metabolism in iPSC-CMs and advise on which metabolic assays to include in future maturation and disease modelin...

Prediction of venous thromboembolism in patients with multiple myeloma treated with lenalidomide, bortezomib, dexamethasone, and transplantation: lessons from the substudy of IFM/DFCI 2009 cohort
CONCLUSIONS: Prospective studies are warranted in light of these results to improve VTE risk stratification and to design adapted thromboprophylaxis in terms of timing and dose.PMID:35557490 | DOI:10.1111/jth.15758
