Dupilumab strengthens HSV-1 specific immune responses in atopic dermatitis
Impaired viral clearance in a subgroup of atopic dermatitis (AD) patients can lead to severe herpes simplex virus (HSV) infections named eczema herpeticum (EH). We recently identified a type 2 skewed viral immune response in EH patients. Clinical data suggest a reduced incidence of EH in AD patients treated with dupilumab, however, immunological investigations of this phenomenon are still lacking.

Dupilumab strengthens herpes simplex virus type 1 –specific immune responses in atopic dermatitis
Atopic dermatitis (AD), one of the most prevalent chronic inflammatory skin diseases, is orchestrated by a complex pathophysiology influenced by the immune system and skin barrier; it is influenced by environmental factors.1 Altered immune response is marked by type 2 inflammation, with a key role for TH2 cells, but it occurs especially in the chronic phase TH1 and TH17 responses as well.2,3 Moderately and severely affected AD patients are particularly prone to bacterial, fungal, and viral infections.

Investigating the Safety and Efficacy of the Synthetic Drug Herbix on Immune Responses Involved in the Treatment of a Mouse Model of Herpes Simplex Virus
In this study, 25 male BALB/c mice were used to conduct a mouse model of herpes by subcutaneously injecting an HSV-1 suspension (100 µL of 1× PFU/mL). The mice were divided into 5 groups with groups 1 to 3 designated as intervention groups, and groups 4 and 5 serving as positive and negative control groups, respectively. After 2 days of virus inoculation, the mice were treated with different concentrations of Herbix (100, 200, and 300 mg/mL) via subcutaneous injection.
