Development of an automated production process of [64Cu][Cu (ATSM)] for positron emission tomography imaging and theranostic applications
Various production methods of [64Cu][Cu (ATSM)] exist, but non-standardized equipment results in varying production qualities often not easily implemented in wider hospital settings. [64Cu][Cu (ATSM)] was synthesized on a modified GE TRACERlab FXN automated synthesis module with high molar activity and good radiostability over 20 h. We demonstrate a simple and feasible synthesis of [64Cu][Cu (ATSM)] based on commercially distributed standardized synthesis modules suitable for PET imaging and theranostic studies of tumor microenvironment hypoxia. Cyclotron-produced copper-64 radioisotope tracers offer the possibility to per...

Novel developed HPLC analyses of [68Ga]Ga/[177Lu]Lu ‐EDTMP and [68Ga]Ga/[177Lu]Lu‐DOTA‐Zoledronate
Practical and realible R-HPLC analyses of [68Ga]Ga/[177Lu]Lu-EDTMP and [177Lu]Lu-DOTA-Zoledronate were exhibited for the first time. Radiolabeled phosphonates have found widespread applications in nuclear medicine for the treatment of bone metastases. Even though considerable attention has been devoted to the administration of radiolabeled phosphonates to the patients, quality control (QC) studies belong to detection of them are still speculative and need to be improved. Herein, practical, efficient, and more reliable high-performance liquid chromatograpy (HPLC) methods were described for the first time. Radiochemical puri...

Automated Radiochemical Synthesis of Pharmaceutical Grade [18F]FLT using 3 ‐N‐Boc‐5’‐O‐dimethoxytrityl‐3’‐O‐nosyl‐thymidine precursor and its Sep‐Pak® Purification employing Selective Elution from Reversed Phase
Pharmaceutical grade 3 ’-deoxy-3’-[18F]fluorothymidine [18F]FLT was synthesized using 3-N-Boc-5 ’-O-dimethoxytrityl-3’-O-nosyl-thymidine (BOC-Nosyl) precursor, in the general purpose TRACERlab FX modules. Purification of [18F]FLT, via solid phase extraction (SPE) after radiosynthesis, using a combination of different SPE cartridges, yielded satisfactory results, with radiochemical and chemical purity> 99%. While the non-decay corrected radiochemical yield (RCY) with 20 mg (24 μmole) of BOC-Nosyl precursor was found to be 6.80 ± 0.16 %, the decay corrected radiochemical yield (RCY) was 9.95 ± 0.24 %, Residual ...

Synthesis of [2H5]baricitinib via [2H5]ethanesulfonyl chloride
Recently, baricitinib has attracted interest for its potential role as a therapeutic agent in the treatment of COVID-19, creating a need to develop a synthetic route to its deuterium-labelled analogue for use as a mass spectrum internal standard, since the isotopologue is typically somewhat costly. This paper presents a new synthetic route to [2H5]baricitinib from [2H5]ethanesulfonyl chloride. Since the preparation and isolation of [2H5]ethanesulfonyl chloride form a major challenge, these comprise a large focus of the overall work. Baricitinib, typically applied as a treatment for rheumatoid arthritis, has recently attrac...

Automated radiosynthesis of the adenosine A2A receptor ‐targeting radiotracer [18F]FLUDA
We report herein a remotely controlled automated radiosynthesis of [18F]FLUDA using a GE TRACERlab FX2 N radiosynthesizer. The radiotracer was obtained by a one-pot two-step radiofluorination procedure (radiochemical yield: 9 ± 1%, radiochemical purity: ≥99%, molar activities: 69–333 GBq/µmol (EOS), total synthesis time: approx. 95 min). Altogether, we successfully established a reliable and reproducible procedure for the automated production of [18F]FLUDA. [18F]FLUDA is a selective radiotracer for in vivo imaging of the adenosine A2A receptor (A2AR) by positron emission tomography (PET). Promising preclinical result...

Automated radiosynthesis of the adenosine A2A receptor ‐targeting radiotracer [18F]FLUDA
[18F]FLUDA is a selective radiotracer for in vivo imaging of the adenosine A2A receptor (A2AR) by positron emission tomography (PET). Promising preclinical results obtained by neuroimaging of mice and piglets suggest the translation of [18F]FLUDA to human PET studies. Thus, we report herein a remotely controlled automated radiosynthesis of [18F]FLUDA using a GE TRACERlab FX2 N radiosynthesizer. The radiotracer was obtained by a one-pot two-step radiofluorination procedure with a radiochemical yield of 9 ± 1%, a radiochemical purity of ≥ 99% and molar activities in the range of 69-333 GBq/μmol at the end of synthesis wi...
