Hippocampal Upregulation of Complement Component C3 in Response to Lipopolysaccharide Stimuli in a Model of Fragile-X Syndrome
Curr Issues Mol Biol. 2023 Nov 18;45(11):9306-9315. doi: 10.3390/cimb45110582.ABSTRACTThe complement system is part of the innate immune system and has been shown to be altered in autism spectrum disorder (ASD). Fragile-X syndrome (FXS) is the main genetic cause of ASD and studies suggest a dysregulation in the immune system in patients with the disorder. To assess if an animal model of FXS presents with altered complement signaling, we treated male Fmr1 knockout (KO) mice with lipopolysaccharide (LPS) and collected the hippocampus 24 h later. Assessment of the expression of the complement genes C1q, C3, and C4 identified ...

Discovery of novel indole derivatives as potent and selective inhibitors of proMMP-9 activation
Bioorg Med Chem Lett. 2023 Nov 10:129541. doi: 10.1016/j.bmcl.2023.129541. Online ahead of print.ABSTRACTMatrix metalloproteinase-9 (MMP-9) is a secreted zinc-dependent endopeptidase that degrades the extracellular matrix and basement membrane of neurons, and then contributes to synaptic plasticity by remodeling the extracellular matrix. Inhibition of MMP-9 activity has therapeutic potential for neurodegenerative diseases such as fragile X syndrome. This paper reports the molecular design, synthesis, and in vitro studies of novel indole derivatives as inhibitors of proMMP-9 activation. High-throughput screening (HTS) of ou...

J Biosci. 2023;48:41.ABSTRACTThe fruit fly, Drosophila melanogaster, has been one of the finest systems for decoding myriad puzzles across different domains of biology. Beyond addressing the fundamental problems, it has been used as a fantastic model organism for human disease research. Being an insect, Drosophila has a robust and advanced olfactory system that has been used many times as a model neuronal circuit to study fundamental questions in neurobiology. The circuit is well-explored at anatomical, physiological, and functional levels. It provides several advantages for the study of neurobiological disorders, such as ...

Altered brain serotonin 5-HT < sub > 1A < /sub > receptor expression and function in juvenile Fmr1 knockout mice
We examined agonist-labeled 5-HT1AR densities in male and female Fmr1 knockout mice, a mouse model of FXS, and found no difference in whole-brain 5-HT1AR expression in adult control compared to Fmr1 knockout mice. However, juvenile Fmr1 knockout mice had significantly lower whole-brain 5-HT1AR expression than age-matched controls. Consistent with these results, juvenile Fmr1 knockout mice showed significantly reduced behavioral responses elicited by the 5-HT1AR agonist (R)-8-OH-DPAT, effects blocked by the selective 5-HT1AR antagonist, WAY-100635. Also, treatment with the selective 5-HT1AR agonist, NLX-112, dose-dependentl...
