Allosteric inhibition of HIV-1 integrase activity. Integrase strand transfer inhibitors (INSTIs), which target the enzyme active site, have witnessed clinical success over the past 5 years, but the generation of drug resistance poses challenges to INSTI-based therapies moving forward. Integrase is a dynamic protein, and its ordered multimerization is critical to enzyme activity. The integrase tetramer, bound to viral DNA, interacts with host LEDGF/p75 protein to tether integration to active genes. Allosteric integrase inhibitors (ALLINIs) that compete with LEDGF/p75 for binding to integrase disrupt integrase assembly with viral DNA and allosterically inhibit enzyme function. ALLINIs display steep dose response curves and synergize with INSTIs ex vivo, highlighting this novel inhibitor class for clinical development. PMID: 23647983 [PubMed - as supplied by publisher]
CONCLUSIONS: This report demonstrates that the streamlined, algorithmic approach effectively identified HIV infections in this large cluster-randomized trial. Longitudinal HIV testing (all participants, all visits) and quality control testing provided useful data on the frequency of errors and provided more accurate data for HIV incidence estimates. ConclusionA comparative high-resolution analysis of B*35Px or B*35Py alleles among healthy controls and HIV+ individuals showed an overrepresentation of HLA-B*35Px alleles in the healthy Indian gene pool (58%) and in the HIV+ cohort (70%). The most common B*35 subtype found among the HIV infected subjects was B*3503/55 (20%; Px group), followed by B*3501 (10%; Py group). Our study concludes that the Indian gene pool possesses higher frequency of B*35Px alleles.
