MAC30 knockdown involved in the activation of the Hippo signaling pathway in breast cancer cells.

This study was carried out to determine the roles and the mechanisms of MAC30 in breast cancer. We used our own data and public database to analyze the MAC30 mRNA and protein levels in breast cancer tissues. In addition, we established MAC30 knockdown breast cancer cells using MAC30 siRNA. The roles of MAC30 were detected by using Soft agar assay, annexinV-FITC/PI double staining, and Transwell assay. Western blotting was used to analyze the potential mechanism(s) of MAC30 in these cells. We found that MAC30 mRNA and protein were higher in the cancer tissues compared to the matched normal tissues. MAC30 expression was associated with tumor size, tumor differentiation and ER status. Overall survival rate of the patients with low MAC30 expression was obviously higher than the ones with high expression. The apoptotic ratio was lower in MDA-MB-231 and MDA-MB-157 cells with MAC30 expression. By Western blot analysis, we found that increased levels of phosphorylated YAP1, MST1, and LATS1 after MAC30 siRNA transfection in these two cells. In summary, we demonstrate that MAC30 knockdown is involved in the activation of the Hippo signaling pathway. PMID: 29990302 [PubMed - as supplied by publisher]

Authors: Zhao Y, Onda K, Sugiyama K, Yuan B, Tanaka S, Takagi N, Hirano T Abstract In the present study, the antitumor effects of arsenic disulfide (As2S2) on the proliferative, survival and migratory ability of human breast cancer MCF‑7 and MDA‑MB‑231 cells were investigated, and its potential underlying molecular mechanisms with an emphasis on cell cycle arrest, apoptosis induction, autophagy induction and reactive oxygen species (ROS) generation were determined. The results indicated that As2S2 significantly inhibited the viability, survival and migration of breast cancer cells in a dose‑dependent m...

Effects of green tea, matcha tea and their components epigallocatechin gallate and quercetin on MCF‑7 and MDA-MB-231 breast carcinoma cells. Oncol Rep. 2018 Oct 12;: Authors: Schröder L, Marahrens P, Koch JG, Heidegger H, Vilsmeier T, Phan-Brehm T, Hofmann S, Mahner S, Jeschke U, Richter DU Abstract We investigated the anticarcinogenic potential of green tea and its components epigallocatechin gallate (EGCG) and quercetin, as well as tamoxifen, on MCF-7 and MDA-MB-23 breast cancer cells. Using high-performance liquid chromatography, the quantity of EGCG and quercetin in green tea was analyzed. T...

Upregulated miR‑203a‑3p and its potential molecular mechanism in breast cancer: A study based on bioinformatics analyses and a comprehensive meta‑analysis. Mol Med Rep. 2018 Oct 10;: Authors: Cai KT, Feng CX, Zhao JC, He RQ, Ma J, Zhong JC Abstract Breast cancer (BC) has been identified as the leading malignancy in women worldwide. However, the potential molecular mechanism of microRNA (miR)‑203a‑3p in BC remains to be elucidated. The present study evaluated the expression of miR‑203a‑3p in BC and adjacent normal tissue in several publically available datasets. The distinguishability of p...

Authors: Petrou P Abstract INTRODUCTION: Breast cancer, the second most common cancer in women, exerts a multidimensional grave effect on affected women. Among the several sub-types of breast cancer, the overexpression of the human epidermal growth factor receptor-2 gene is associated with poorer survival and higher relapse rates. Trastuzumab, a monoclonal antibody, has proved its safety and efficacy in this specific subtype, nevertheless its high cost urges for caution among payers. Areas covered: The scope of this study is to systematically search and critically assess the economic evaluation studies of trastuzum...
