Enhancement of mouse hematopoietic stem/progenitor cell function via transient gene delivery using integration-deficient lentiviral vectors.
• Integration-deficient vectors (IdLV) express genes transiently in dividing stem cells• Hematopoietic stem/progenitor (HSPC) cells can be programmed using IdLV• HOXB4 or Angptl3 expression from IdLV improves engraftment of transplanted HSPC• Short-term gene delivery avoids side effects asso ciated with constitutive expression• Disabled HIV has been modified so it cannot facilitate integration of thei delivered genome, causing short-term gene expression in dividing cells. Used in stem cells, this can alter their properties; for example, to improve the efficiency of bone marrow transpl antation.
