815 T helper 2 cell immunity eliminates pre-cancerous skin lesions

Actinic keratosis (AK) is a precursor to cutaneous squamous cell carcinoma (SCC). Effective therapeutics to eliminate AKs and prevent SCC are lacking. Long treatment durations and severe side effects have limited the efficacy of cytotoxic AK treatments including 5-fluorouracil (5-FU) chemotherapy and photodynamic therapy. Previously, we have shown that thymic stromal lymphopoietin (TSLP), an epithelium-derived cytokine, induces a robust antitumor immunity in the skin. Since this discovery, we have demonstrated the high efficacy of calcipotriol, a topical TSLP inducer, in combination with 5-fluorouracil (5-FU) as a novel immunotherapy for AK treatment with potential for SCC prevention.
