Downregulation of hsa_circ_0007534 restricts the proliferation and invasion of cervical cancer through regulating miR-498/BMI-1 signaling
Publication date: Available online 21 August 2019Source: Life SciencesAuthor(s): Xuan Rong, Wei Gao, Xiaomei Yang, Junjun GuoAbstractAimsHsa_circ_0007534 has been reported to be a novel cancer-related circRNA affecting multiple cancers. However, little is known about the role of hsa_circ_0007534 in cervical cancer specifically. In the current study, we aimed to explore the expression and function of hsa_circ_0007534 in cervical cancer.Main methodsThe expression of circRNA, miRNA and mRNA was measured using real-time quantitative polymerase chain reactions. Protein expression was assessed by Western blot. Knockdown of has_c...

Honokiol induces endoplasmic reticulum stress-mediated apoptosis in human lung cancer cells
Publication date: Available online 29 January 2019Source: Life SciencesAuthor(s): Jianfei Zhu, Shuonan Xu, Wenli Gao, Jianyu Feng, Guolong ZhaoAbstractAimsHonokiol is a hydroxylated biphenyl natural product and displays potent antitumor activity against several cancers including prostate cancer, melanoma, leukemia, and colorectal cancer. The present study was to investigate the in vitro activity of honokiol against A549 and 95-D human lung cancer cells.Main methodsA549 and 95-D cells were used with honokiol treatment. Cell viability was determined by CCK-8 assay. The cell migration and apoptosis were evaluated by wound hea...

Inhibition of HOXB7 suppresses p27-mediated acute lymphoblastic leukemia by regulating basic fibroblast growth factor and ERK1/2
In this study, we explored the molecular mechanism of HOXB7 in cell viability and cell cycle in ALL cell lines.Materials and methodsPeripheral blood lymphocytes was isolated by Isopycnic Ficoll-Hypaque solution; Relative mRNA expression of HOXB7 was measured by RT-qPCR; Relative protein expressions of HOXB7, p27, bFGF, pERK1/2 were tested by Western blot assay; Cell viability was tested by MTT; Cell proliferation was detected by BrdU assay; 2.8. Cell cycle was analyzed by flow cytometry.Key findingsHOXB7 was significantly elevated in peripheral blood lymphocytes of patients with ALL. HOXB7 was inhibited by HOXB7 siRNA tran...

Osteopontin plays a unique role in resistance of CD34+/CD123+ human leukemia cell lines KG1a to parthenolide
Conclusion Parthenolide in spite of the reduction in gene expression of AKT, mTOR or beta-catenin, stimulates the OPN expression in KG1a cells. The OPN expression pattern in KG1a cells could be compatible with CD34+/CD123+ subtype enrichment by PTL which in turn implies OPN's unique role in resistance of cell populations characterized by CD34+/CD123+ phenotype.

Acquired expression of osteopontin selectively promotes enrichment of leukemia stem cells through AKT/mTOR/PTEN/β-catenin pathways in AML cells
Publication date: 1 May 2016 Source:Life Sciences, Volume 152 Author(s): Saeed Mohammadi, Seyed H. Ghaffari, Mojgan Shaiegan, Mahin Nikougoftar Zarif, Mohsen Nikbakht, Shiva Akbari Birgani, Kamran Alimoghadam, Ardeshir Ghavamzadeh Aims Acute myeloid leukemia (AML) initiation and progression have been attributed to subpopulations of self-renewing leukemia stem cells (LSCs), which contribute to progression, recurrence and therapeutic resistance in leukemia. Osteopontin (OPN) plays an important role in promoting survival and drug resistance in LSCs. The aim of this study was to explore OPN roles in modulating curcum...
