Molecules, Vol. 27, Pages 6236: Akebia Saponin D Inhibits the Inflammatory Reaction by Inhibiting the IL-6-STAT3-DNMT3b Axis and Activating the Nrf2 Pathway
This study aimed to preliminarily explore the anti-inflammatory effect of ASD and the underlying mechanisms from the perspective of DNA methylation and inflammation-related pathways. We found that ASD significantly reduced the production of multiple inflammatory mediators, including nitric oxide (NO) and prostaglandin E2 (PGE2), in LPS-induced RAW264.7 cells. The expression of DNA methyltransferase (DNMT) 3b and inducible nitric oxide synthase (iNOS) was also obviously inhibited by the ASD treatment. The protein and mRNA levels of interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) were als...

Molecules, Vol. 27, Pages 1990: Dihydrohomoplantagin and Homoplantaginin, Major Flavonoid Glycosides from Salvia plebeia R. Br. Inhibit oxLDL-Induced Endothelial Cell Injury and Restrict Atherosclerosis via Activating Nrf2 Anti-Oxidation Signal Pathway
In this study, we evaluated the effect of homoplantaginin and its derivative dihydrohomoplantagin on oxLDL-induced endothelial cell injury and atherosclerosis in apoE-/- mice. Our results showedthat both dihydrohomoplantagin and homoplantaginin inhibited apoptosis and the increased level of ICAM-1 and VCAM-1 in oxLDL-stimulated HUVECs and the plaque endothelium of apoE-/- mice. Additionally, both of them restricted atherosclerosis development of apoE-/- mice. Mechanistic studies showed that oxLDL-induced the increase in ROS production, phosphorylation of ERK and nuclear translocation of NF-κB in HUVECs was si...

Potential inhibitors and plant based technology: An alternative approach to treat corona virus
Bioorg Chem. 2021 Nov 1;117:105460. doi: 10.1016/j.bioorg.2021.105460. Online ahead of print.ABSTRACTThe current pneumonia outbreak, which began in early December 2019 near Wuhan City, Hubei Province, China, is caused by a novel corona virus (CoV) known as '2019-nCoV' or '2019 novel corona virus or COVID-19' by the World Health Organization (WHO). Vaccines are available to prevent corona virus contagious infection or to reduce the viral load in body but virus is continuously mutating itself to infect people at severity. In this critical scenario this review provide a compiled study for techniques and tools that can be used...

Molecules, Vol. 25, Pages 4591: Magnolol Enhances the Therapeutic Effects of TRAIL through DR5 Upregulation and Downregulation of c-FLIP and Mcl-1 Proteins in Cancer Cells
Kwon Magnolol is a biologically active compound, isolated from the Chinese herb Magnolia, that regulates antiproliferative, anticancer, antiangiogenic and antimetastatic activities. We found that magnolol sensitizes TRAIL-induced apoptotic cell death via upregulation of DR5 and downregulation of cellular FLICE-inhibitory protein (c-FLIP) and Mcl-1 in cancer cells, but not in normal cells. Mechanistically, magnolol increased ATF4-dependent DR5 expression at the transcription level, and knockdown of ATF4 markedly inhibited magnolol-induced DR5 upregulation. Silencing DR5 with siRNA prevented combined treatment with magn...

Preparation of poly(glutamic acid) shielding micelles self-assembled from polylysine-b-polyphenylalanine for gene and drug codelivery
Publication date: Available online 19 February 2020Source: Chinese Chemical LettersAuthor(s): Jing Ma, Jingpeng Zhang, Lin Chi, Chong Liu, Yanhui Li, Huayu TianABSTRACTA novel amphiphilic cationic block copolymer polylysine-b-polyphenylalanine (PLL-b-PPhe) was synthesized and self-assembled into micelles in aqueous solution, then shielded with poly(glutamic acid) (marked as PG/PLL-b-PPhe) to codeliver gene and drug for combination cancer therapy. Here, doxorubicin (DOX) was selected to be loaded into PLL-b-PPhe micelles and the drug loading efficiency was 8.0%. The drug release studies revealed that the PLL-b-PPhe micelles...

A supramolecular co-delivery strategy for combined breast cancer treatment and metastasis prevention
Publication date: Available online 13 June 2019Source: Chinese Chemical LettersAuthor(s): Yuxuan Chen, Bowen Li, Xiaohong Chen, Min Wu, Yongtao Ji, Guping Tang, Yuan PingAbstractWe herein propose a co-delivery approach where small interference RNA (siRNA) and anticancer chemotherapeutic drug are simultaneously loaded into a single delivery carrier for the combined treatment of breast cancer and metastasis prevention. The co-delivery vector is composed of chondroitin sulfate (CS)-coated β-cyclodextrin-polyethylenemine polymer, which is capable of loading paclitaxel (PTX) and siRNA simultaneously to form therapeutic nanocom...

A reactive oxygen species-responsive dendrimer with low cytotoxicity for efficient and targeted gene delivery
Publication date: Available online 25 March 2019Source: Chinese Chemical LettersAuthor(s): Yueqi Wang, Changjian Li, Libo Du, Yang LiuAbstractNonviral vectors have been attracting more attention for several advantages in gene delivery and the development of nonviral gene carriers with high delivery efficiency and low cytotoxicity has long been a key project. Starburst polyamidoamine dendrimers are a class of synthetic polymers with unique structural and physical characteristics. However, when they are used as gene carrier, the gene transfection efficiency is not satisfactory. Herein, a novel thioketal-core polyamidoamine d...

ROS-induced HSP70 promotes cytoplasmic translocation of high-mobility group box 1b and stimulates antiviral autophagy in grass carp kidney cells Cell Biology
In this study, we found that both grass carp reovirus (GCRV) infection and hydrogen peroxide (H2O2) treatment induced the accumulation of reactive oxygen species (ROS) in C. idella kidney cells and stimulate autophagy. Suppressing ROS accumulation with N-acetyl-l-cysteine significantly inhibited GCRV-induced autophagy activation and enhanced GCRV replication. Although ROS-induced autophagy, in turn, restricted GCRV replication, further investigation revealed that the multifunctional cellular protein high-mobility group box 1b (HMGB1b) serves as a heat shock protein 70 (HSP70)–dependent, pro-autophagic protein in grass ca...

Advances in quantum dot-mediated siRNA delivery
Publication date: Available online 13 July 2017 Source:Chinese Chemical Letters Author(s): Zhi-Yao He, Zhao-Hui Jin, Mei Zhan, Zhou Qin, Zhi Li, Ting Xu Nano-sized quantum dots (QDs) exhibit uniquely optical properties that are tunable with different sizes and shapes. QDs can emit narrow symmetric bands under a wide excitation range, possess anti-photobleaching stability, and be bio-functionalized on the large surface area. Therefore, QDs are attractive vectors for imaging-guided therapy. Small-interfering RNA (siRNAs)-based therapeutics hold great potential to target a large part of the currently undruggable genes, but o...
