Antibody ‐Functionalized Gold Nanostar‐Mediated On‐Resonance Picosecond Laser Optoporation for Targeted Delivery of RNA Therapeutics
In this study, an elaborated plasmonic optoporation approach is proposed to efficiently and selectively transfect specific cells. The site ‐specific optoporation is obtained by tuning the spectral range of a supercontinuum pulsed picosecond laser in order for each individual cell binding gold nanostar with their unique resonance peak to magnify the local field strength in the near‐infrared region and facilitate a selective delivery of small interfering RNA, messenger RNA, and Cas9‐ribonucleoprotein into human retinal pigment epithelial cells. Numerical simulations indicate that optoporation is not due to a plasma‐m...

Targeted Combination of Antioxidative and Anti ‐Inflammatory Therapy of Rheumatoid Arthritis using Multifunctional Dendrimer‐Entrapped Gold Nanoparticles as a Platform
A multifunctional dendrimer nanoplatform is developed to have entrapped gold nanoparticles within dendrimer interiors, and covalently linked PEGylated alpha ‐tocopheryl succinate, PEGylated folic acid, and 1,3‐propane sultone on dendrimer peripheries. The designed nanoplatform enables co‐delivery of drug/gene to macrophage cells, effectively scavenging reactive oxygen species and inhibiting tumor necrosis factor‐α expression for combined antiox idative and anti‐inflammatory therapy of rheumatoid arthritis. AbstractAbundant reactive oxygen species and tumor necrosis factor ‐α (TNF‐α) cytokine supply of M1...

Quantitative Assessment of Endosomal Escape of Various Endocytosed Polymer ‐Encapsulated Molecular Cargos upon Photothermal Heating
A methodology is reported to monitor the dynamic process of 43 fluorescent cargos released from the polymeric capsules and the lysosomes where capsules are trapped, and their spread in the cytosol upon short laser irradiation. Intracellular spread cannot be described by free diffusion, but is determined by interaction of the molecular cargo with intracellular components. AbstractEncapsulated molecular cargos are efficiently endocytosed by cells. For cytosolic delivery, understanding the dynamic process of cargos release from the carrier vehicles used for encapsulation and the lysosomes where the carrier vehicles are trappe...

Assembling Defined DNA Nanostructure with Nitrogen ‐Enriched Carbon Dots for Theranostic Cancer Applications
Noncanonical self ‐assembly of DNA is an attractive strategy to expand their applications in multidisciplinary fields and is rarely explored. This work reports a nitrogen‐enriched carbon dots (NCDs)‐mediated DNA self‐assembly strategy. NCDs‐conjugated DNA nanostructures may be a potential theranostic platfo rm with further fine‐tuned NCDs properties such as near‐red fluorescence or photothermal activities. AbstractDNA nanostructures as scaffolds for drug delivery, biosensing, and bioimaging are hindered by its vulnerability in physiological settings, less favorable of incorporating arbitrary guest molecules a...

Biodegradable Nanocarriers Resembling Extracellular Vesicles Deliver Genetic Material with the Highest Efficiency to Various Cell Types
This work demonstrates a new tool for the transfer of genetic material. The developed biodegradable polyelectrolyte nanocapsules exhibit high levels of uptake and transfer to various cell types, including hematopoietic stem cells. A co ‐transfer and regulated release of several types of RNAs or DNAs are possible using this approach, and are able to revolutionize gene transfer technology. AbstractEfficient delivery of genetic material to primary cells remains challenging. Here, efficient transfer of genetic material is presented using synthetic biodegradable nanocarriers, resembling extracellular vesicles in their biomech...

Sequential Targeting TGF ‐β Signaling and KRAS Mutation Increases Therapeutic Efficacy in Pancreatic Cancer
Fraxinellone ‐loaded CGKRK‐modified nanoparticles are prepared for regulating transforming growth factor beta signals in the suppressive tumor microenvironment. Based on this, the dense stroma is modulated and the intratumor blood perfusion is enhanced, meaning the delivery barrier of the following siRNA ‐loaded lipid‐coated calcium phosphate (LCP) biomimetic high‐density lipoprotein nanoparticles (siKras‐LCP‐ApoE3) are internalized and specifically silence KRAS mutation and kill cancer cells. AbstractPancreatic cancer is a highly aggressive malignancy that strongly resists extant treatments. The failure of e...
