GSE234780 Knockdown TIMM13 in A549 lung cancer cell line
Contributors : Zhaozhao Zhao ; Ting NiSeries Type : Expression profiling by high throughput sequencingOrganism : Homo sapiensWe depleted TIMM13 using short interfering RNA (siRNA) in A549 lung cancer cells, and found that silencing of TIMM13 could significantly inhibit A549 cell proliferation. Thus, we conducted RNA-seq in the A549 cells transduced with either nontargeting siRNA or two distinct TIMM13-specific siRNAs. We observed widespread gene expression change in A549 cells upon TIMM13 knockdown.

GSE161226 HEDGEHOG/GLI Modulates The PRR11-SKA2 Bidirectional Transcription Unit in Lung Squamous Cell Carcinomas
Contributors : Yiyun Sun ; Ying ZhangSeries Type : Expression profiling by high throughput sequencingOrganism : Homo sapiensWe previously demonstrated that proline-rich protein 11 (PRR11) and spindle and kinetochore associated 2 (SKA2) constituted a head-to-head gene pair driven by a prototypical bidirectional promoter, and this gene pair synergistically promoted the development of non-small-cell lung cancer. However, the signaling pathways leading to the ectopic expression of this gene pair remain obscure. In the present study, we first analyzed the lung squamous cell carcinoma (LSCC) relevant RNA sequencing data from the...

GSE66606 Gene expression profiling of Calu-6 lung cancer cells transfected with scrambled siRNA and LAPTM4B-specific siRNA
Contributors : Yuho Maki ; Humam KadaraSeries Type : Expression profiling by arrayOrganism : Homo sapiensWe recently characterized the adjacent airway field of cancerization in NSCLC by whole transcriptome expression analysis and demonstrated that lysosomal protein transmembrane 4 beta (LAPTM4B) was an elevated field cancerization marker in NSCLCs and in adjacent but not in distant normal-appearing airways. We also found that LAPTM4B was up-regulated in NSCLCs compared to normal lung and promoted anchorage-dependent growth of lung cancer cells. Previous reports suggested that LAPTM4B is activated following metabolic and gen...

GSE81727 Cordycepin induces apoptosis by caveolin-1 mediated JNK’s regulation of Foxo3a in lung cancer A549 cells
Contributors : Jung M Kim ; Ik-Soon JangSeries Type : Expression profiling by arrayOrganism : Homo sapiensc-Jun N-terminal kinase (JNK) plays a pivotal role in the regulation of cancer cell apoptosis. Previous studies have revealed that forkhead transcription factor (Foxo3a) is a critical effector of JNK-mediated tumor suppression. However, it is not clear whether caveolin-1 (CAV1) mediated JNK/Foxo3a pathway is involved in cancer cell apoptosis. We found that cordycepin upregulates CAV1 expression, which was accompanied by JNK phosphorylation (p-JNK), which induced Foxo3a translocation into the nucleus, resulting in the u...

GSE66604 Inhibition of ABCB1 Overcomes Cancer Stem Cell-like Properties and Acquired Resistance to MET inhibitor in Non-Small Cell Lung Cancer
In this study, we tried to identify the mechanism of acquired resistance to MET inhibitor. We analyzed the antitumor effects of two MET inhibitors, PHA-665752 and crizotinib, in 10 NSCLC cell lines. EBC1 cells with MET amplification were the only cells that were sensitive to both MET inhibitors. We established PHA-665752-resistant EBC1 cells, namely EBC1-R cells. EBC1-R cells showed overexpression of ATP-binding cassette sub-family B member 1 (ABCB1) as well as phosphorylation of MET. EBC1-R cells grew as cell spheres that exhibited cancer stem cell-like (CSC) properties and epithelial mesenchymal transition (EMT). The lev...
