CARD9 mediates glucose-stimulated insulin secretion in pancreatic beta cells
Biochem Pharmacol. 2021 Jul 4:114670. doi: 10.1016/j.bcp.2021.114670. Online ahead of print.ABSTRACTCaspase recruitment domain containing protein 9 (CARD9) plays key regulatory role(s) in innate and adaptive immune responses. Recent evidence implicates CARD9 in the onset of metabolic diseases including insulin resistance. However, potential contributory roles of CARD9 in glucose-stimulated insulin secretion (GSIS) remain unknown. Herein, we report that CARD9 is expressed in human islets, rat islets, mouse islets and clonal INS-1 832/13 cells. Subcellularly, CARD9 is predominantly cytosolic (∼75%) in INS 1- 832/13 cells. ...

Cynandione A and PHA-543613 inhibit inflammation and stimulate macrophageal IL-10 expression following α7 nAChR activation
Biochem Pharmacol. 2021 May 13:114600. doi: 10.1016/j.bcp.2021.114600. Online ahead of print.ABSTRACTCynandione A, an acetophenone isolated from Cynanchum Wilfordii Radix, attenuates inflammation. The present study aimed to study the mechanisms underlying cynandione A-induced antiinflammation. Treatment with cynandione A and the specific α7 nicotinic acetylcholine receptor (α7 nAChR) agonist PHA-543613 remarkably reduced overexpression of proinflammatory cytokines, including tumor necrosis factor (TNF)-α, interleukin (IL)-6 and IL-1β in lipopolysaccharide (LPS)-treated RAW264.7 cells and primary peritoneal macrophages,...

Overexpression of retinoid X receptor beta provides protection against oxidized low-density lipoprotein-induced inflammation via regulating PGC1 α-dependent mitochondrial homeostasis in endothelial cells
Biochem Pharmacol. 2021 Apr 16:114559. doi: 10.1016/j.bcp.2021.114559. Online ahead of print.ABSTRACTRetinoid X receptor beta (RXRβ) has been poorly studied in atherosclerosis. The aim of the present study is to explore the function of RXRβ in oxidized low density lipoprotein (ox-LDL)-induced inflammation in endothelial cells and the underlying mechanism. The protein expression of RXRβ in the aorta of atherosclerotic mice was detected. A lentivirus vector for RXRβ overexpression and RNA interference for RXRβ downregulation were constructed and transfected into human aortic endothelial cells (HAECs). The results showed...

Tetrahydroxystilbene glycoside improves endothelial dysfunction and hypertension in obese rats: the role of omentin-1
CONCLUSIONS: Down-regulation of omentin-1 induces endothelial dysfunction and hypertension in obesity. TSG treatment (at least partially) increases omentin-1 via promoting binding of PPAR-γ and Itln-1 promoter in adipose tissues, subsequently exerts protective effects on endothelial function via activating Akt/eNOS/NO signaling and attenuating oxidative/nitrative stress. These results suggest that TSG could be developed as a promising anti-hypertension agent that protects against endothelial dysfunction and obesity-associated cardiovascular diseases.PMID:33647262 | DOI:10.1016/j.bcp.2021.114489

Resveratrol enhances transintestinal cholesterol excretion through selective activation of intestinal liver X receptor alpha
In conclusion, RSV could decrease circulating cholesterol levels through enhancing TICE and limiting cholesterol absorption via selective activation of intestinal LXRα.PMID:33631191 | DOI:10.1016/j.bcp.2021.114481
