Polysaccharide-based (kappa carrageenan/carboxymethyl chitosan) nanofibrous membrane loaded with antifibrinolytic drug for rapid hemostasis- in vitro and in vivo evaluation
Int J Biol Macromol. 2023 Jul 12:125786. doi: 10.1016/j.ijbiomac.2023.125786. Online ahead of print.ABSTRACTThis work aimed to establish a novel membrane consisting of hemostatic polysaccharides, kappa-carrageenan (KC), and carboxymethyl chitosan (CMC) in tandem with polyvinyl alcohol that spun together as a matrix and loaded with tranexamic acid (TXA) as antifibrinolytic agent for further coagulation effect during and after oral surgeries. The electrospinning of KC was done for the first time and in comparison of CMC has better hemostatic efficacy. The effect of the hemostat was investigated by its surface morphology (SEM...

Biomimetic and Multifunctional Hemostatic Hydrogel with Rapid Thermoresponsive Gelation and Robust Wet Adhesion for Emergency Hemostasis: A Rational Design Based on Photo-Cross-Linking Coordinated Hydrophilic-Hydrophobic Balance Strategies
Biomacromolecules. 2023 Jun 27. doi: 10.1021/acs.biomac.3c00357. Online ahead of print.ABSTRACTUncontrolled bleeding in emergency situations is a great threat to both military and civilian lives, and an ideal hemostat for effectively controlling prehospital hemorrhage is urgently needed but still lacking. Although hemostatic hydrogels are promising for emergency hemostasis, they are currently challenged by either the mutual exclusion between a short gelation time and strong adhesive network or the insufficient functionality of ingredients and complicated operations for in situ curing. Herein, an extracellular matrix biopol...

Electrospun kaolin-loaded chitosan/PEO nanofibers for rapid hemostasis and accelerated wound healing
In this study, the classic hemostatic chitosan and the most well-known inorganic hemostatic agent-kaolin were tried to combine to form a more excellent dressing. Together with the aid of non-toxic, harmless and good hydrophilic polyethylene oxide, chitosan/polyethylene oxide (PEO)/kaolin nanofiber membranes (CPKs) were prepared by electrospinning technology. Such membranes exhibited adjustable mechanical properties and good biocompatibility. Furthermore, a series of in vitro coagulation experiments proved that CPKs with 10 % ratio of kaolin (CPK10) has excellent hemostatic ability. Especially, in the whole blood coagulatio...

In-vitro and in-vivo hemostat evaluation of decellularized liver extra cellular matrix loaded chitosan/gelatin spongy scaffolds for liver injury
Int J Biol Macromol. 2021 Oct 25:S0141-8130(21)02286-8. doi: 10.1016/j.ijbiomac.2021.10.128. Online ahead of print.ABSTRACTIndividually, Chitosan (C) and Gelatin (G) are increasingly being used for the simulation and testing of surgical procedures. In the present study, at combination of chitosan/gelatin (CG) was optimized and later enriched by the loading decellularized liver extracellular matrix powder (dLECM) prepared from porcine liver, we hypothesized CG-dLECM combination would enhance wound healing and reduce postoperative complications after liver surgery. Varying concentration of dLECM (1, 4, and 8 mg/ml) were load...

Physico-mechanical and biological evaluation of an injectable m-TG cross-linked thrombin loaded amended gelatin hemostat to heal liver trauma
The objective of this study was to evaluate the activation of a coagulation system both in vitro and in vivo. We assessed detailed physical characteristics of a microbial transglutaminase (m-TG) crosslinked thrombin (TB) laden Gelatin (Gel) hemostat sealant in vitro and its hemostatic efficacy for controlling bleeding caused by liver trauma in rats as well as its efficacy for organ regeneration after making a critical defect. The prepared hemostat gel showed almost seven times higher absorbance behavior than a negative control. Thrombogenicity of the prepared gel was determined based on platelet adhesion, whole blood clott...

Targeting Biological Polyanions in Blood: Strategies towards the Design of Therapeutics.
Abstract In this review, we highlight well-described and emerging polyanions, and the way these molecules can be targeted in the design of potential therapeutics (synthetic and biologics) with applications in thrombosis and hemostasis. It is important to strike a balance between bleeding and clotting. In thrombosis, unwanted blood clots are formed in the lumen of a blood vessel, obstructing the flow of blood through the circulatory system. Over many years of research, several polyanionic biopolymers that can either impede (anticoagulant) or promote (procoagulant) blood clotting have been identified. Mediators impe...

Efficient antibacterial dextran-montmorillonite composite sponge for rapid hemostasis with wound healing.
Abstract Montmorillonite (MMT) powder, as the most effective hemostats in natural silicates, is restricted for commercial application due to its embolic effect. Until now, it's still a challenge to control the leakage of MMT and avoid its side-effects. Herein, poly aldehyde dextran (PDA)/MMT composite sponge (PM) with commendable tissue adhesion, antibacterial, and wound healing performances is developed for massive hemorrhage control. Based on the high degree of perfect synergism of PDA and MMT, the PM sponge can rapidly seal the wound, and promote cells aggregation and adhesion, whole coagulation system activati...

Rational design of porous starch/hyaluronic acid composites for hemostasis.
Abstract Effective hemorrhage control is pivotal for decreasing the trauma death both in civilian and military but has proven to be dauntingly challenging, especially for solid viscera and artery trauma. Here we report the fabrication of a novel starch-based hemostat, sodium trimethaphosphate (STMP)-crosslinked starch/hyaluronic acid (HA) (ScSH) porous composites. Aiming at hemostatic potential, physicochemical properties, cytocompatibility, hemocompatibility, histocompatibility and hemostatic performance of ScSH composites have been studied. As it turned out, the incorporation of HA greatly improved the water abs...

Comparison of acid and water-soluble chitosan doped fibrous cellulose hemostat wet laid nonwoven web for hemorrhage application.
Abstract Fibrous hemostat was produced using wet laying technique comprising of acid and water soluble chitosan with fibrous cellulose. The hemostatic efficiency and their structural properties were evaluated using blood clotting time, whole blood adhesion and analytical characterization techniques. The maximum concentration of chitosan utilised for the production of nonwoven was 1.5 w/v % upon which increased the viscosity of the solution. The absorption of phosphate buffer solution was found to be 1117% for acid soluble chitosan based nonwoven whereas for water soluble chitosan based nonwoven was 997%. Increase...
