Development and Validation of a Genomic Profile for the Omission of Local Adjuvant Radiation in Breast Cancer
CONCLUSION: The novel POLAR genomic signature on the basis of LRR biology may identify patients with a low risk of LRR despite not receiving RT, and thus may be candidates for RT omission.PMID:36599119 | DOI:10.1200/JCO.22.00655

In conclusion, circulating monocytes in older adults exhibit increased expression of activation, adhesion, and migration markers, but decreased expression of co-inhibitory molecules. MERTK Inhibition Increases Bone Density via Increased Osteoblast Activity https://www.fightaging.org/archives/2022/12/mertk-inhibition-increases-bone-density-via-increased-osteoblast-activity/ Bone density results from the balance of constant activity on the part of osteoblasts and osteoclasts, the former building bone, the latter breaking it down. With advancing age, the balance of activity shifts to favor osteoclasts, prod...

Doping with aromatase inhibitors and oestrogen receptor modulators in steroid users: Analysis of a forum to identify dosages, durations and adverse drug reactions
CONCLUSIONS: Patient narratives posted on forums can be a useful tool in the context of doping, to better understand practices, motivations and possibly to bring up pharmacovigilance signals.PMID:35660259 | DOI:10.1016/j.therap.2022.03.004

Exercise Causes Arrhythmogenic Remodeling of Intracellular Calcium Dynamics in Plakophilin-2-Deficient Hearts
Conclusions: Exercise disproportionately affects Ca2+i homeostasis in PKP2-deficient hearts in a manner facilitated by stimulation of intracellular β-adrenergic receptors and hyperphosphorylation of phospholamban. These cellular changes create a proarrhythmogenic state that can be mitigated by ryanodine receptor 2 blockade. Our data unveil an arrhythmogenic mechanism for exercise-induced or catecholaminergic life-threatening arrhythmias in the setting of PKP2 deficit. We suggest that membrane-permeable β-blockers are potentially more efficient for patients with arrhythmogenic right ventricular cardiomyopathy, highlight t...
