Multiplex genotyping of SNPs in genomic DNA via hydrogel-based assay mediated with MutS and polyethylene glycol
In this study, a hydrogel-based multiplex SNP genotyping platform specifically designed for genomic DNA is presented. This platform integrates the ligation detection reaction (LDR) and rolling circle amplification (RCA) techniques within a hydrogel-based multiplex sensing system, enabling adaptable and sensitive SNP genotyping for genomic DNA. To enhance the specificity of the assay, MutS protein and polyethylene glycol are introduced into the protocol, reducing the non-specific ligation and RCA reactions synergistically. With significant specificity improvement of over 10-fold, three types of SNPs within an artificially c...

Loratadine oral bioavailability enhancement via solid dispersion loaded oro-dispersible films: Formulation, characterization and pharmacokinetics
Colloids Surf B Biointerfaces. 2023 Aug 25;230:113526. doi: 10.1016/j.colsurfb.2023.113526. Online ahead of print.ABSTRACTLoratadine (LRD) belongs to second-generation tricyclic H1 antihistamine class, known for its non-sedating properties in allergic reactions. H1 antihistamines avoid and block the responses to allergens or histamine in nose and conjunctivae, thereby abolishing itching, congestion and sneezing. LRD is a Biopharmaceutical Class System (BCS) class II drug with dissolution or solubility limited absorption which limited the oral bioavailability and therapeutic efficacy of LRD. To improve the oral bioavailabil...

Polyethylene glycol modified graphene oxide-silver nanoparticles nanocomposite as a novel antibacterial material with high stability and activity
Colloids Surf B Biointerfaces. 2023 Jun 30;229:113435. doi: 10.1016/j.colsurfb.2023.113435. Online ahead of print.ABSTRACTInorganic antibacterial nanomaterials play an increasingly important role in addressing the growing threat of drug-resistant bacteria. Graphene oxide-silver nanoparticles composite (GO-AgNPs), as a kind of inorganic nanomaterials, have excellent antibacterial properties, showing promising potential in biomedical field. However, GO-AgNPs are terribly prone to sedimentation due to aggregation in physiological solutions, along with its non-environmental issues during the synthesis process, seriously limits...

Targeted paclitaxel prodrug nanoassemblies to improve therapeutic effects for liver cancer
Colloids Surf B Biointerfaces. 2023 Mar 30;226:113285. doi: 10.1016/j.colsurfb.2023.113285. Online ahead of print.ABSTRACTProdrug nanoassemblies fabricated by anticancer drug conjugates exhibited more advantages in controlled drug release and bioavailability and favorable antitumor efficacy. In this paper, lactobionic acid (LA) was connected with polyethylene glycol through amido linkages, and paclitaxel was joined with polyethylene glycol by means of ester bonds to form the prodrug copolymer LA-PEG-PTX. Then, LA-PEG-PTX was automatically assembled into LA-PEG-PTX nanoparticles (LPP NPs) by dialysis. The LPP NPs had a rela...

Intracellular trafficking kinetics of nucleic acid escape from lipid nanoparticles via fluorescence imaging
CONCLUSION: Here, we demonstrated the ability to track uptake and the sub-cellular fate of LNPs containing pDNA and mRNA, enabling improved screening prior to in vivo studies which would aid in formulation optimization.PMID:37016519 | DOI:10.2174/1389201024666230403094238

Multifunctional electrospun membranes with hydrophilic and hydrophobic gradients property for wound dressing
Colloids Surf B Biointerfaces. 2023 Mar 24;225:113276. doi: 10.1016/j.colsurfb.2023.113276. Online ahead of print.ABSTRACTAchieving sustained and stable release of macromolecular antibacterial agents and unidirectional transport of liquids in targeted environment is still a challenge to be addressed in the management of wounds with large amounts of tissue exudates. In this work, a multilayer electrospun membrane (ethylcellulose-ethylcellulose/gelatin-quercetin/Eudragit L-100/polyethylene glycol, EC-EC/Gel-Q/EL/PEG) was designed with hydrophobic-hydrophilic gradients and drug sustained-release properties controlled by self-...

L-asparaginase immobilization in supramolecular nanogels of PEG-grafted poly HPMA and bis( α-cyclodextrin) to enhance pharmacokinetics and lower enzyme antigenicity
Colloids Surf B Biointerfaces. 2023 Mar 1;225:113234. doi: 10.1016/j.colsurfb.2023.113234. Online ahead of print.ABSTRACTL-asparaginase (ASNase) enzyme has limited therapeutic use due to its poor pharmacokinetics and immunogenicity. To overcome these obstacles, we immobilized ASNase in biocompatible poly hydroxypropyl methacrylamide (P(HPMA))-based nanogels simply formed through the host-guest inclusion complex of ASNase-conjugated random copolymer of HPMA and polyethylene glycol (PEG) acrylate (P(HPMA-MPEGA)) and α-cyclodextrin dimer (bisCD) using cystamine as a linker. The effects of bisCD and polymer concentrations on ...
