Quantitative profiling of genes associated with cancer pathways in brain tumors
CONCLUSION: Based on the obtained data, we can conclude that individual diagnoses differ in transcriptomic profile. An individual molecular approach is therefore necessary in order to provide comprehensive and targeted therapy on multiple metabolic pathways in the diagnosis of brain tumors.PMID:37353350 | DOI:10.48095/ccko2023224

Cocrystallization of lenvatinib and temozolomide to improve the performance in stability, dissolution, and tabletability
CrystEngComm, 2023, Accepted Manuscript DOI: 10.1039/D3CE00473B, PaperZhi-Qing Wang, Xia-Lin Dai, Dai-Lin Gu, Chao Wu, Tongbu Lu, Xiangtian Long, Jia-Mei Chen Two anti-melanoma drugs, lenvatinib and temozolomide, were cocrystallized to obtain two drug-drug cocrystal solvates, TMZ-LEN ·MeOH (1:1:1) and TMZ-LEN·EtOH (1:1:1). They were fully characterized by XRD and thermal analyses, NMR and... The content of this RSS Feed (c) The Royal Society of Chemistry

A Click Chemistry-Based Artificial Metallo-Nuclease
Angew Chem Int Ed Engl. 2023 Jun 20:e202305759. doi: 10.1002/anie.202305759. Online ahead of print.ABSTRACTArtificial metallonucleases (AMNs) are promising DNA damaging drug candidates. Here, we demonstrate how the 1,2,3-triazole linker produced by the copper-catalysed azide-alkyne cycloaddition (CuAAC) reaction can be directed to build copper-binding AMN scaffolds. We selected biologically inert reaction partners tri-azidomethyl-mesitylene and ethynyl-thiophene to develop TC-Thio, a bioactive C3-symmetric ligand in which three thiophene-triazole moieties are positioned around a central mesitylene core. The ligand was char...

Therapeutic Responses to Combination Nivolumab and Temozolomide as Salvage Therapy for Metastatic Melanoma: A Case Series
Oncologist. 2023 Jun 20:oyad184. doi: 10.1093/oncolo/oyad184. Online ahead of print.ABSTRACTThe management of metastatic melanoma patients that fail multiple lines of systemic therapy remains a significant challenge. There is limited literature regarding combination of anti-PD-1 and temozolomide, or of other chemotherapy agents, in melanoma. Here, we present a series of 3 patients with metastatic melanoma and their responses to nivolumab and temozolomide combination therapy after progression on several local/regional therapies, combination immune checkpoint inhibitors, and/or targeted therapies. The novel combinatory strat...

Bavituximab decreases immunosuppressive myeloid-derived suppressor cells in newly diagnosed glioblastoma patients
DISCUSSION: Bavituximab has activity in newly diagnosed GBM and resulted in on-target depletion of intratumoral immunosuppressive MDSCs. Elevated pre-treatment expression of myeloid-related transcripts in GBM may predict response to bavituximab.PMID:37327319 | DOI:10.1158/1078-0432.CCR-23-0203

CCL21-CCR7 signaling promotes microglia/macrophage recruitment and chemotherapy resistance in glioblastoma
Cell Mol Life Sci. 2023 Jun 14;80(7):179. doi: 10.1007/s00018-023-04788-7.ABSTRACTGlioblastoma (GBM) is the most common and fatal primary tumor of the central nervous system (CNS) and current treatments have limited success. Chemokine signaling regulates both malignant cells and stromal cells of the tumor microenvironment (TME), constituting a potential therapeutic target against brain cancers. Here, we investigated the C-C chemokine receptor type 7 (CCR7) and the chemokine (C-C-motif) ligand 21 (CCL21) for their expression and function in human GBM and then assessed their therapeutic potential in preclinical mouse GBM mod...

HOXD-AS2-STAT3 feedback loop attenuates sensitivity to temozolomide in glioblastoma
CONCLUSION: Our study elucidated the crucial role of the HOXD-AS2-STAT3 positive feedback loop in regulating TMZ sensitivity, suggesting that this could be provided as a potential therapeutic candidate of glioblastoma.PMID:37308741 | DOI:10.1111/cns.14277
