TNFAIP3 may be key to TLR4-activation of the inflammasome in the retinal vasculature
Exp Eye Res. 2022 May 11:109108. doi: 10.1016/j.exer.2022.109108. Online ahead of print.ABSTRACTThe goal of these studies were to determine whether tumor necrosis factor, alpha-induced protein 3 (TNFAIP3) regulated toll-like receptor 4 (TLR4) actions on the NOD-like receptor protein 3 (NLRP3) inflammasome. Western blotting was done on retinal lysates from TLR4 floxed and endothelial cell specific TLR4 knockout mice for TNFAIP3, TLR4, and NLRP3 pathway proteins. Retinal endothelial cells (REC) were grown in normal (5mM) and high glucose (25mM) and treated with TNFAIP3 siRNA, followed by Western blotting for TLR4 and NLRP3 p...

Inhibition of Drp1 ameliorates diabetic retinopathy by regulating mitochondrial homeostasis
Exp Eye Res. 2022 Apr 28:109095. doi: 10.1016/j.exer.2022.109095. Online ahead of print.ABSTRACTDiabetic retinopathy (DR) is a potentially blinding complication resulting from diabetes mellitus (DM). Retinal vascular endothelial cells (RMECs) dysfunction occupies an important position in the pathogenesis of DR, and mitochondrial disorders play a vital role in RMECs dysfunction. However, the detailed mechanisms underlying DR-induced mitochondrial disorders in RMECs remain elusive. In the present study, we used High glucose (HG)-induced RMECs in vitro and streptozotocin (STZ)-induced Sprague-Dawley rats in vivo to explore th...

Regulation of inflammation by VEGF/BDNF signaling in mouse retinal M üller glial cells exposed to high glucose
We examined the expression of glutamine-synthetase (GS), glial fibrillary acidic protein (GFAP), vascular-endothelial growth factor (VEGF), interleukin-1beta (IL-1β), and tumor necrosis factor-alpha (TNF-α) at different time points after mouse retinal MGCs exposed to high glucose (25 mM). We also explored changes in the expression of brain-derived neurotrophic factor (BDNF), nuclear factor kappa B (NF-κB), IL-1β, and TNF-α in MGCs after treatments with anti-VEGF, VEGF siRNA, BDNF siRNA, BDNF recombination protein, and NF-κB inhibitor. In mouse retinal MGCs exposed to high glucose, BDNF was increased after treatments ...

A transcriptomics and molecular biology based investigation reveals the protective effect and mechanism of carnosol on t-BHP induced HRMECs via Nrf2 signaling pathway
In this study, carnosol was found to inhibit HRMECs injury induced by t-BHP. Transcriptomics and molecular biology illustrated that the mechanism was associated with oxidative stress, vascular system development, apoptosis, cell cycle, cell adhesion, cytoskeleton, and nitric oxide biosynthesis. Carnosol directly scavenged free radicals or activated the Nrf2 signaling pathway to alleviate HRMECs oxidative stress. ML385 pretreatment or Nrf2 small interference RNA (siRNA) inhibited the protective effect of carnosol on HRMECs injury. Moreover, the apoptosis and cell cycle arrest in HRMECs were suppressed by carnosol. Treatment...

C-terminal binding protein 2 promotes high-glucose-triggered cell proliferation, angiogenesis and cellular adhesion of human retinal endothelial cell line
ConclusionThis study implicates that CtBP2 promotes HG-induced cell proliferation, angiogenesis and cellular adhesion, and CtBP2 might be a potential target in the prevention of DR.

Novel Role of Prereplication Complex Component Cell Division Cycle 6 in Retinal Neovascularization
CONCLUSIONS: The above observations clearly reveal that PLCβ3-mediated NFATc1 activation-dependent CDC6 expression plays a crucial role in VEGFA/oxygen-induced retinopathy-induced retinal neovascularization.PMID:35236105 | DOI:10.1161/ATVBAHA.121.317182
