PMA-triggered PKCε activity enhances Nrf2-mediated antiviral response on fish rhabdovirus infection
Publication date: April 2019Source: Fish & Shellfish Immunology, Volume 87Author(s): Yun-Fei Dang, Tian-Xiu Qiu, Da-Wei Song, Lei LiuAbstractViral infection is often accompanied with alteration of intracellular redox state, especially an imbalance between reactive oxygen species (ROS) production and antioxidant cellular defenses. The previous studies showed that an antioxidant cellular defense system, the transcription factor nuclear factor erythroid 2-related factor 2 (Nrf2), played an important role against spring viraemia of carp virus (SVCV) infection in fish. To further reveal the mediated mechanism that Nrf2 active s...

Publication date: April 2019Source: Redox Biology, Volume 22Author(s): Daniel S. de Jesus, Evan DeVallance, Yao Li, Micol Falabella, Danielle Guimaraes, Sruti Shiva, Brett A. Kaufman, Mark T. Gladwin, Patrick J. PaganoAbstractPulmonary arterial hypertension (PAH) is a complex degenerative disorder marked by aberrant vascular remodeling associated with hyperproliferation and migration of endothelial cells (ECs). Previous reports implicated bone morphogenetic protein antagonist Gremlin 1 in this process; however, little is known of the molecular mechanisms involved. The current study was designed to test whether redox signal...

Renalase attenuates mitochondrial fission in cisplatin-induced acute kidney injury via modulating sirtuin-3
In this study, we showed that the protective effect of recombinant renalase in cisplatin-induced AKI was associated with the regulation of ROS production, mitochondrial dynamics and sirtuin-3 (Sirt3) levels in vivo and in vitro. After cisplatin treatment, recombinant renalase restored Sirt3 expression, reduced mitochondrial fission and ROS generation. In HK-2 cells, downregulation of endogenous Sirt3 expression by siRNA transfection abrogated the renalase cytoprotection.SignificanceOur study suggests that renalase protects against cisplatin-induced AKI by improving mitochondrial function and inhibiting oxidative stress, an...

Translocator protein 18 kDa ligand alleviates neointimal hyperplasia in the diabetic rat artery injury model via activating PKG
This study suggests that TSPO inhibition suppresses the proliferation and migration of VSMCs induced by hyperglycemia, consequently, preventing atherosclerosis and restenosis after angioplasty in diabetic conditions. TSPO may be a potential therapeutic target to reduce arterial remodeling induced by angioplasty in diabetes.

The role of the BMP4/Smad1 signaling pathway in mesangial cell proliferation: A possible mechanism of diabetic nephropathy
This study explored the role of the BMP4/Smad1 signaling pathway in mesangial matrix expansion during the process of diabetic nephropathy.Main methodsDiabetic rats were induced by high-fat feeding followed by an intraperitoneal injection of streptozotocin. Glomerular lesions were examined. Immunohistochemical analysis was performed in order to identify BMP4/Smad1 signaling proteins (BMP4, ALK3, and Smad1) and mesangial ECM proteins (Col1 and Col4) in kidney tissue. Cell proliferation and the expression of BMP4, Smad1 and Col4 were determined in cultured mesangial cells exposed to high glucose. The specific regulatory role ...

Telmisartan attenuates diabetic nephropathy progression by inhibiting the dimerization of angiotensin type-1 receptor and adiponectin receptor-1
This study examined the effect of telmisartan on diabetic nephropathy (DN) and its underlying mechanism.Main methodsDiabetes was induced in rats through a single intraperitoneal injection of streptozotocin. Diabetic rats treated with or without the intravenous injection of AdipoR1 siRNA were intragastrically administered with 5 mg/kg/d telmisartan or a vehicle for 12 weeks. The rat proximal tubular epithelial cell line NRK-52E was treated with HG (30 mmol/L) with or without telmisartan (10 μM) for 48 h.Key findingsIn streptozotocin-induced diabetic rats, telmisartan treatment could decrease the inulin clearance ...

LRP1 activation attenuates white matter injury by modulating microglial polarization through Shc1/PI3K/Akt pathway after subarachnoid hemorrhage in rats
Publication date: Available online 23 January 2019Source: Redox BiologyAuthor(s): Jianhua Peng, Jinwei Pang, Lei Huang, Budbazar Enkhjargal, Tongyu Zhang, Jun Mo, Pei Wu, Weilin Xu, Yuchun Zuo, Jun Peng, Gang Zuo, Ligang Chen, Jiping Tang, John H. Zhang, Yong JiangAbstractWhite matter injury (WMI) is associated with motor deficits and cognitive dysfunctions in subarachnoid hemorrhage (SAH) patients. Therapeutic strategy targeting WMI would likely improve the neurological outcomes after SAH. Low-density lipoprotein receptor-related protein-1 (LRP1), a scavenger receptor of apolipoprotein E (apoE), is able to modulate microg...

Molecular cloning and preliminary functional analysis of six RING-between-ring (RBR) genes in grass carp (Ctenopharyngodon idellus)
Publication date: April 2019Source: Fish & Shellfish Immunology, Volume 87Author(s): Lifei Luo, Denghui Zhu, Rong Huang, Lv Xiong, Rumana Mehjabin, Libo He, Lanjie Liao, Yongming Li, Zuoyan Zhu, Yaping WangAbstractUbiquitination is a post-translational modification of proteins that is widely present in eukaryotic cells. There is increasing evidence that ubiquitinated proteins play crucial roles in the immune response process. In mammals, RING-between-RING (RBR) proteins play a key role in regulating immune signaling as the important E3 ubiquitin ligases during ubiquitination. However, the function of RBR in fish is still u...

Inhibiting Inflammation and Modulating Oxidative Stress in Oxalate-Induced Nephrolithiasis with the Nrf2 Activator Dimethyl Fumarate.
This study aimed to investigate whether nuclear factor (erythroid-derived 2)-like 2 (Nrf2, also called Nfe2l2) induced by dimethyl fumarate (DMF) could protect renal epithelial cells against oxalate-mediated injury both in vivo and in vitro. Glyoxylic acid monohydrate was intraperitoneally injected into Sprague-Dawley rats with or without intragastric administration of DMF. We showed that calcium oxalate crystallisation, accompanied by overexpression of oxidant species and inflammatory cytokines and apoptosis in the rat kidney, was partially reversed by treatment with DMF. Furthermore, oxalate induced a reduction in cell v...

Specific Delivery of Delta-5-Desaturase siRNA via RNA Nanoparticles Supplemented with Dihomo-γ-Linolenic Acid for Colon Cancer Suppression
In this study, we employed RNA nanotechnology for specific delivery of D5D-siRNA to xenograft colon tumors using 3WJ RNA nanoparticles. When a targeting module, i.e., the EpCAM aptamer, was incorporated, the 3WJ pRNA nanoparticles were able specifically deliver D5D siRNA to human colon cancer HCA-7 cells both in vitro and in vivo, resulting in significant downregulation of D5D expression. Co-treatment with DGLA in combination with 3WJ-EpCAM-siRNA induced a higher DGLA/AA ratio and enhanced formation of 8-HOA at a threshold level, and in HCA-7 tumor-bearing mice, induced significant tumor suppression. We further confirmed t...
