Silencing of Gal-7 inhibits TGF- β1-induced apoptosis of human airway epithelial cells through JNK signaling pathway.
In this study, we investigated the expression and role of Gal-7 in the apoptosis of bronchial epithelial cells BEAS-2B upon TGF-β1 stimulation. TGF-β1 significantly induced apoptosis of BEAS-2B cells, as determined by flow cytometry. Western blot results revealed that the mRNA and protein expression of Gal-7 were obviously increased after TGF-β1 stimulation. Small interfering RNA (siRNA)-mediated knockdown of Gal-7 abrogated TGF-β1-evoked cell apoptosis. Simultaneously, increased Bcl-2 expression, decreased Bax expression and the cleavage of poly ADP-ribose polymerase (PARP) and caspase-3 activity were also monitored i...

Store-operated Calcium Entry-associated Regulatory Factor Regulates Airway Inflammation and Airway Remodeling in Asthma Mice Models
CONCLUSIONS: SARAF plays a protective role against airway inflammation and remodeling in asthma mice models by blunting SOCE; SARAF may also be a functional regulating factor of hASMCs.PMID:34231388 | DOI:10.1152/ajplung.00079.2020

Amphiregulin induces CCN2 and fibronectin expression by TGF- β through EGFR-dependent pathway in lung epithelial cells
CONCLUSION: These results suggested that AREG mediates the TGF-β-induced EMT in human lung epithelial cells through EGFR/JNK/AP-1 activation. Understanding the role of AREG in the EMT could foster the development of therapeutic strategies for airway remodeling in severe asthma.PMID:36578010 | DOI:10.1186/s12931-022-02285-2

Ephedrine induced Thioredoxin-1 expression through β-adrenergic receptors/Cyclic AMP/protein kinase A/dopamine- and Cyclic AMP-regulated phosphoprotein signaling pathway.
In this study, we found that Eph induced Trx-1 expression, which was inhibited by propranolol (β-adrenergic receptors inhibitor), but not by phenoxybenzamine (α-adrenergic receptors inhibitor) in rat pheochromocytoma PC12 cells. Moreover, the increase of Trx-1 expression was inhibited by SQ22536 (adenylyl cyclase inhibitor) and H-89 (protein kinase A inhibitor). Interestingly, the effect of Eph on dopamine- and Cyclic AMP-regulated phosphoprotein (DARPP-32) was similar to Trx-1. Thus, the relationship between Trx-1 and DARPP-32 was further studied. The DARPP-32 siRNA significantly reduced Trx-1 expression, but Trx-1 siRN...

The MNK-1/eIF4E pathway as a new therapeutic pathway to target inflammation and remodelling in asthma.
Abstract Therapeutic targets in asthma are reduction of airway inflammation and remodelling, the latter is not affected by available drugs. Here we present data that inhibition of MAPK-activated protein kinase (MNK)-1 reduces inflammation and remodelling. MNK-1 regulates protein expression by controlling mRNA stability, nuclear export and translation through the eukaryotic initiation factor 4E (eIF4E). Airway smooth muscle cells were derived from asthmatic and non-asthmatic donors. Cells were pre-treated with CGP57380 (MNK-1 inhibitor) or MNK-1 siRNA, before TNF-α stimulation. Cytokine and protein expression was ...

IgE production in CD40/CD40L cross-talk of B and mast cells and mediator release via TGase 2 in mouse allergic asthma.
This study aimed to investigate the role of TGase 2 on IgE production and signaling pathways in mast cell activation related to OVA-induced allergic asthma. Bone marrow-derived mast cells (BMMCs) isolated from WT or TGase 2(-/-) mice were activated with Ag/Ab (refer to act-WT-BMMCs and act-KO-BMMCs, respectively). B cells isolated from splenocytes were activated with anti-mouse IgM (act-B cells), and B cells were co-cultured with BMMCs. WT and TGase 2(-/-) mice were sensitized and challenged with OVA adsorbed in alum hydroxide. Intracellular Ca(2+) ([Ca(2+)]i) levels were determined by fluorescence intensity; IgE, mediator...

Human airway smooth muscle cells secrete amphiregulin via bradykinin/COX-2/PGE2, inducing COX-2 ,CXCL8 and VEGF expression in airway epithelial cells.
Abstract Human airway smooth muscle cells (HASMC) contribute to asthma pathophysiology through an increased smooth muscle mass and elevated cytokine/chemokine output. Little is known about how HASMC and the airway epithelium interact to regulate chronic airway inflammation and remodelling. Amphiregulin is a member of the family of epidermal growth factor receptor (EGFR) agonists with cell growth and pro-inflammatory roles and increased expression in the lungs of asthma patients. Here we show that bradykinin (BK) stimulation of HASMC increases amphiregulin secretion in a mechanism dependent upon BK induced COX-2 ex...

Endothelin-1 Induces VCAM-1 Expression-Mediated Inflammation via Receptor Tyrosine Kinases and Elk/p300 in Human Tracheal Smooth Muscle Cells.
Abstract The elevated level of endothelin-1 (ET-1) has been detected in the bronchoalveolar lavage of patients with severe asthma, acute lung injury, acute respiratory distress syndrome and sepsis. ET-1 may affect vessel tone together with lung physiology and pathology. Vascular cell adhesion molecule-1 (VCAM-1), is one kind of adhesion molecules participating in the process of polymorphonuclear leukocytes transmigration and regulating the occurrence and amplification of tissue inflammation. However, the molecular mechanisms underlying ET-1 mediated expression of VCAM-1 on human tracheal smooth muscle cells (HTSMC...

IFNγ-induced JAK/STAT-, but not NF-κB-, signaling pathway is insensitive to glucocorticoid in airway epithelial cells.
Abstract Although the majority of asthmatics are well controlled by inhaled glucocorticoids (GCs), severe asthmatics are poorly responsive to GCs. This latter group is responsible for a disproportionate share of healthcare costs associated with asthma. Recent studies in immune cells have incriminated interferon gamma (IFNγ) as a possible trigger of GC insensitivity in severe asthma; however, little is known about the role of IFNγ in modulating GC effects in other clinically relevant non-immune cells, such as airway epithelial cells. We hypothesized that IFNγ-induced JAK/STAT-associated signaling pathways in air...
