CRISPR/Cas9-mediated deletion of adipocyte genes associated with NAFLD alters adipocyte lipid handling and reduces steatosis in hepatocytes < em > in vitro < /em >
Am J Physiol Cell Physiol. 2023 Sep 18. doi: 10.1152/ajpcell.00291.2023. Online ahead of print.ABSTRACTObesity is a major risk factor for the development of non-alcoholic fatty liver disease (NAFLD) and the subcutaneous white adipose tissue (scWAT) is the primary lipid storage depot and regulates lipid fluxes to other organs. Our previous work identified genes up-regulated in scWAT of patients with NAFLD: SOCS3, DUSP1, and SIK1. Herein, we knocked down (KD) their expression in human adipose-derived mesenchymal stem cells (hADMSCs) using CRISPR/Cas9 technology and characterized their phenotype. We found that SOCS3, DUSP1, a...

The ALKBH5/SOX4 axis promotes liver cancer stem cell properties via activating the SHH signaling pathway
J Cancer Res Clin Oncol. 2023 Aug 30. doi: 10.1007/s00432-023-05309-6. Online ahead of print.ABSTRACTHepatocellular carcinoma (HCC), featured with high prevalence and poor prognosis, is the major cause of cancer-related deaths worldwide. As a subgroup of liver cancer cells capable of differentiation, tumorigenesis and self-renewal, liver cancer stem cells (LCSCs) serve as one of the reasons leading to HCC progression and therapeutic resistance. Therefore, in-depth exploration of novel molecular biomarkers related to LSCSs is of great necessity. In our study, we found that human AlkB homolog H5 (ALKBH5) expression was enric...

Acyl-CoA Medium-Chain Synthetase-3 (ACSM3) Is Regulated by Insulin Like Growth Factor 2 mRNA Binding Protein 3 (IGF2BP3) and Inhibits Proliferation, Motility and Stem Cell Properties of Breast Invasive Carcinoma
CONCLUSION: We found that ACSM3 was regulated by IGF2BP3 and attenuated BRCA proliferation, invasion and stem cell properties. The role of ACSM3 in BRCA was first revealed, which provides a novel target for treatment.PMID:37625834
