Development and functional diversification of dendritic cells in immunity
Immunology Interest Group Among many seminal discoveries, Dr Murphy’s work provided the first demonstration in vivo of antigen-induced apoptosis in the thymus during thymocyte selection and identified mechanisms by which pathogens instruct the direction of T helper differentiation. His work also revealed the basis for TH2 stability as a result of extinction of IL-12 signaling and transcriptional auto-activation. More recently Dr. Murphy's research addresses the regulation of immune responses with particular emphasis on dendritic cell (DC) differentiation, central to both effective defense and rheumatologic disorders. Hi...

NIH Research Festival: A Celebration of Intramural Science Plenary Session III: Chronic Inflammation
Most of the NIH institutes and centers are involved in studies for which chronic inflammation is a common denominator. Allergies, asthma, heart and circulatory disease, rheumatoid arthritis, inflammatory bowel disease, celiac disease, some cancers, multiple sclerosis, and possibly Alzheimer’s. These are just a few debilitating diseases that can originate partly or entirely from the immune system’s aggressive response to perceived harmful stimuli. Aging itself has been described as the totality of a lifetime of chronic inflammation. Better understanding the causes and control of chronic inflammation thus can lead to pro...

Neutrophils in the pathogenesis of systemic autoimmune diseases: casting the NET widely
NIH Director's Seminar Series Systemic autoimmune diseases comprise a clinically heterogeneous group of disorders characterized by a failure in self-tolerance to a wide variety of intracellular autoantigens, and include conditions like systemic lupus erythematosus, rheumatoid arthritis and anti-neutrophil cytoplasmic antibody associated-vasculitis. Work at the Systemic Autoimmunity Branch has focused on characterizing the role that aberrant neutrophils play in the promotion of loss of immunological tolerance, amplification of inflammatory responses and direct end-organ damage in these diseases. One characteristic of ...
