Doctors issue new treatment guidelines for skin abscesses caused by MRSA
It has been more than 10 years since the clinical battle began with community-acquired methicillin-resistant Staphylococcus aureus (MRSA), and doctors are still grappling with how to diagnose, treat and prevent this virulent form of staph infection, which is immune to many antibiotics.   As MRSA cases have increased dramatically over the decade, so have the number of skin abscesses — generally pus-filled boils or pimples with discharge — that characterize these infections. Now, researchers from UCLA have issued updated guidelines outlining the best ways to treat and manage these abscesses.   The ...

Treatment of Infections Due to Resistant Staphylococcus aureus
This chapter reviews data on the treatment of infections caused by drug-resistant Staphylococcus aureus, particularly methicillin-resistant S. aureus (MRSA). This review covers findings reported in the English language medical literature up to January of 2013. Despite the emergence of resistant and multidrug-resistant S. aureus, we have seven effective drugs in clinical use for which little resistance has been observed: vancomycin, quinupristin–dalfopristin, linezolid, tigecycline, telavancin, ceftaroline, and daptomycin. However, vancomycin is less effective for infections with MRSA isolates that have a higher MIC w...

Extrachromosomal Inducible Expression
Inducible expression systems are very convenient for proteins that induce strong side effects such as retardation of growth or development and are essential for the expression of toxic proteins. In this chapter we describe the doxycycline-inducible expression system, optimized for the controlled expression in. Two types of inducible plasmids are presented, in which transcription is induced by either adding or removing doxycycline, respectively. Detailed protocols are provided for the construction of the plasmids and the inducible expression of the target protein. (Source: Springer protocols feed by Microbiology)

Target Validation in Mice by Constitutive and Conditional RNAi
We describe three strategies to control gene silencing in mice that can be applied to any transcript of interest. This shRNA based approach enables either i) constitutive body-wide knockdown, ii) cell type-specific knockdown controlled by Cre recombinase, or iii) inducible body-wide knockdown controlled by doxycycline. For reliable expression the shRNA vector of interest is inserted into a Rosa26 docking site of ES cells by a site-specific recombinase. These ES cells can then be used to generate shRNA transgenic mice. This technology enables the production of adult knockdown mice within 11 months for an expedite in vivo va...
