Epigenetic regulation has profound influence on stem cell fate during normal development in maintenance of physiologic tissue homeostasis. Here we report diminished ten-eleven translocation (TET) methylcytosine dioxygenase expression and loss of the DNA hydroxymethylation mark, 5-hydroxymethylcytosine (5-hmC), in keratinocyte stem cells (KSCs) and transit-amplifying cells (TACs) in human psoriasis and in imiquimod-induced murine psoriasis. Loss of 5-hmC was associated with dysregulated KSC kinetics, resulting in accumulation of nestin and FABP5-expressing TACs to produce classic psoriatic epidermal architecture.

In this study, we investigated the factor(s) involved. We found that stanniocalcin‐1 (STC1), a cytokine , is expressed at the basal layer of epidermis. Knockdown of STC1 with siRNA in HaCaT cells decreased matrix metalloproteinase 1 (MMP1) expression, suggesting that STC1 serves as an autocrine factor, maintaining MMP1 mRNA expression in the epidermal layer. Keratin 17(K17), as a cytoskeletal protein, is highly expressed in epithelial cells of inflammatory skin diseases and various tumor tissues. In recent years, the present of K17 in the nucleus was found in several tumors. However, such phenomenon had not been documented in inflammatory dermatoses. In our present study, K17 translocation into the nucleus of keratinocytes in psoriasis were found. Thus, greater insight into the mechanism of K17 translocation and its pathogenicity were explored. Initially, we found that K17 occurs as discrete punctate and/or in a diffuse pattern in the nucleus of keratinocytes in psoriastic lesion.
