In Silico Models for Drug Resistance
We present two in silico models for the discovery of drug resistance mechanisms and for combating the evolution of resistance, respectively. In the first model, we computationally investigated subgraphs of a biological interaction network that show substantial adaptations when cells transcriptionally respond to a changing environment or treatment. As a case study, we investigated the response of the malaria parasite Plasmodium falciparum to chloroquine and tetracycline treatments. The second model involves a machine learning technique that combines clustering, common distance similarity measurements, and hierarchical clust...

Environmental Engineers Seek Strategy To Control Antibiotic Resistant Genes
Rice University researchers "cured" a strain of bacteria of its ability to resist an antibiotic in an experiment that has implications for a long-standing public health crisis. Rice environmental engineer Pedro Alvarez and his team managed to remove the ability of the Pseudomonas aeruginosa microorganism to resist the antibiotic medication tetracycline by limiting its access to food and oxygen... Antibiotic-resistant diseases pose 'apocalyptic' threat, top expert says
Chief medical officer Dame Sally Davies tells MPs issue should be added to national risk register of civil emergenciesBritain's most senior medical adviser has warned MPs that the rise in drug-resistant diseases could trigger a national emergency comparable to a catastrophic terrorist attack, pandemic flu or major coastal flooding.Dame Sally Davies, the chief medical officer, said the threat from infections that are resistant to frontline antibiotics was so serious that the issue should be added to the government's national risk register of civil emergencies. She described what she called an "apocalyptic scenario" where peo...

Identification of Pathogenetically Relevant Genes in Lymphomagenesis by shRNA Library Screens
RNA interference (RNAi) is a conserved posttranscriptional gene silencing mechanism that has recently emerged as a breakthrough genetic tool in functional genomics and drug target discovery. An increasing number of studies applying RNAi in high-throughput screens have begun to unravel complex signaling networks underlying diverse cellular processes. This chapter describes an approach to construct a conditional small-hairpin (sh)RNA library and its application in human lymphoma cell lines. A library cloning procedure outlines the incorporation of shRNA sequences and random 60-mer “bar code” oligonucleotides, ena...

Cholera and Diarrhea
CHOLERA and DIARRHEA At this very moment there is an outbreak of cholera in Zimbabwe– not a worldwide pandemic, but a localized epidemic. This is not a trivial point of interest, but a major news event that will not only have a dramatic impact on the population of Zimbabwe but also on the entire world as the aftermath of this epidemic will require international aid. Cholera will most likely kill thousands of people in Zimbabwe, impact tens of thousands of lives, and will continue to be a constant threat around the world and a very real concern for the adventurous and international travelers. What is cholera? Cholera...
