Tissue hypoxia during ischemic stroke: Adaptive clues from hypoxia-tolerant animal models.
We describe specific physiological and molecular adaptations employed by different animals' models of hypoxia tolerance in aquatic and terrestrial environments. We highlight how these adaptations might provide potential clues on strategies to adapt for the clinical management of tissue hypoxia during conditions such as stroke where oxygen demand fails to match the supply. Silencing lncRNA HOTAIR improves the recovery of neurological function in ischemic stroke via the miR-148a-3p/KLF6 axis
In this study, we explored the effect and potential molecular mechanism of lncRNA homeobox antisense non-coding RNA (HOTAIR) in IS. Permanent middle cerebral artery occlusion (pMCAO) model and oxygen and glucose deprivation (OGD) model were established. HOTAIR was increased in vivo and in vitro models post-ischemic. HOTAIR knockdown promoted neurological function recovery, manifesting in decreased modified neurological severity score, cerebral infarcted area, apoptosis and inflammation, and improved balance ability, spatial learning and memory ability. Abstract There is cumulative evidence that the serine-threonine kinase Akt and its downstream nuclear transcription factor CREB are involved in neuronal survival and protection. Diabetes impairs spatial learning and memory and hippocampal neurogenesis via BDNF in rats with transient global ischemia.
Abstract Diabetic conditions worsen the prognosis of stroke. The molecular mechanism underlying the impairment of post-stroke recovery is not very clear. Here, we establish a rat model resembling human cerebral infarction with or without diabetes to determine how diabetes impairs cognitive recovery. Our data show that diabetes inhibits hippocampal BDNF expression and impairs the survival and differentiation of the newborn neural cells in rats with ischemia.
