Circulating Extracellular Vesicle-Propagated microRNA Signature as a Vascular Calcification Factor in Chronic Kidney Disease
CONCLUSIONS: The miRNA transcriptomic signature of circulating sEVs uncovered their pathologic role, devoid of the calcification-protective miRNAs that target VEGFA signaling in CKD-driven vascular calcification. These sEV-propagated miRNAs are potential biomarkers and therapeutic targets for vascular calcification.PMID:36700539 | DOI:10.1161/CIRCRESAHA.122.321939

Orai1 Inhibitors as Potential Treatments for Pulmonary Arterial Hypertension
CONCLUSIONS: In human PAH and experimental PH, Orai1 expression and activity are increased. Orai1 inhibition normalizes the PAH-hPASMCs phenotype and attenuates PH in rat models. These results suggest that Orai1 should be considered as a relevant therapeutic target for PAH.PMID:36164973 | DOI:10.1161/CIRCRESAHA.122.321041

GSK-3 β Localizes to the Cardiac Z-Disc to Maintain Length Dependent Activation
CONCLUSIONS: We identified a novel mechanism by which GSK-3β localizes to the myofilament to modulate LDA. Importantly, z-disc GSK-3β levels were reduced in patients with heart failure, indicating z-disc localized GSK-3β is a possible therapeutic target to restore the Frank-Starling mechanism in patients with heart failure.PMID:35168370 | DOI:10.1161/CIRCRESAHA.121.319491

Novel Mechanisms of Exosome-Mediated Phagocytosis of Dead Cells in Injured Heart
Conclusions: Our data demonstrates that exosome-associated MFGE8 on one hand enhances opsonization of dead cells and on the other activates phagocytic signaling thus augmenting removal of apoptotic cells, resolution of inflammation and therefore efficient cardiac recovery after injury.PMID:34623174 | DOI:10.1161/CIRCRESAHA.120.317900

Growth Arrest Specific-6 and Axl Coordinate Inflammation and Hypertension
Conclusions: We define a previously unrecognized interaction of human endothelial cells and monocytes that promote formation of AS DCs in hypertension and show a critical role of GAS6 and Axl signaling in both immune cells and endothelial cells. This pathway is potentially a novel therapeutic target to reduce inflammation and end organ damage in hypertension.PMID:34565181 | DOI:10.1161/CIRCRESAHA.121.319643
