STRA6 promotes thyroid carcinoma progression via activation of the ILK/AKT/mTOR axis in cells and female nude mice
CONCLUSIONS: Our study demonstrates the critical roles of STRA6 contributing to TC progression via the ILK/AKT/mTOR axis, which may provide a novel prognostic marker as well as a promising therapeutic target for aggressive TC.PMID:36592123 | DOI:10.1210/endocr/bqac215

MARK2 and MARK4 regulate Sertoli cell BTB dynamics through microtubule and actin cytoskeletons
Endocrinology. 2022 Aug 12:bqac130. doi: 10.1210/endocr/bqac130. Online ahead of print.ABSTRACTMARKs (microtubule affinity-regulating kinases) are non-receptor Ser/Thr protein kinases known to regulate cell polarity and microtubule dynamics in C. elegans, Drosophila, invertebrates, vertebrates and mammals. An earlier study has shown that MARK4 is present at the ectoplasmic specialization (EC) and blood-testis barrier (BTB) in the seminiferous epithelium of adult rat testes. Here, we report the function of MARK4 and another isoform MARK2 in Sertoli cells at the BTB. Knockdown of MARK2, MARK4 or MARK2 and MARK4 by RNAi using...

PCP protein inversin regulates testis function through changes in cytoskeletal organization of actin and microtubules
Endocrinology. 2022 Feb 1:bqac009. doi: 10.1210/endocr/bqac009. Online ahead of print.ABSTRACTInversin is an integrated component of the Frizzled (Fzd)/Dishevelled (Dvl)/Diversin planar cell polarity (PCP) complex that is known to work in concert with the Van Gogh-like protein (e.g., Vangl2)/Prickle PCP complex to support tissue and organ development including the brain, kidney, pancreas and others. However, with the exception of Dvl3 and Vangl2, other PCP proteins have not been investigated in the testis. Herein, we used the technique of RNAi to examine the role of inversin (Invs) in Sertoli cell and testis function by co...

Long Noncoding RNA MIAT Modulates the Extracellular Matrix Deposition in Leiomyomas by Sponging MiR-29 Family
The objective of this study was to determine the expression and functional role of a long noncoding RNA (lncRNA) MIAT (myocardial infarction associated transcript) in leiomyoma pathogenesis. Leiomyoma as compared with myometrium (N=66) expressed significantly more MIAT which was independent of race/ethnicity and menstrual cycle phase but dependent on MED12 (mediator complex subunit 12) mutation status. Leiomyomas bearing the MED12 mutation expressed higher levels of MIAT and lower levels of microRNA 29 family (miR-29a, -b and -c) compared with MED12 wild type leiomyomas. Using luciferase reporter activity and RNA immunopre...

LH induces de novo cholesterol biosynthesis via SREBP activation in granulosa cells during ovulation in female mice
In this study, when immature mice were treated with gonadotropins (eCG followed by hCG), eCG induced and hCG maintained the expression of SREBP-1a, -2, and SCAP granulosa cells, whereas INSIG1 expression was dramatically downregulated after hCG injection. Downregulation of INSIG1 led to generate the SREBPs active form and translocate the SREBPs active form to nuclei. Inhibition of generation of the SREBPs active form by fatostatin or Scap siRNA in both in vivo and in vitro significantly decreased the expressions of de novo cholesterol biosynthetic enzymes, cholesterol accumulation, and progesterone (P4) production compared...
