GSE120512 Transcriptome modulation by loss-of-YAP1/TAZ in SK-N-MC Ewing Sarcoma cells
Contributors : Juan Diaz-Martin ; Laura Romero-PerezSeries Type : Expression profiling by arrayOrganism : Homo sapiensYAP1 (Yes-associated protein 1) and TAZ (transcriptional coactivator with PDZ-binding motif, or WWTR1) are nucleo-cytoplasmic shuttling proteins that can function in the nucleus as transcriptional coactivators. Here we sought to evaluate which genes are regulated by endogenous levels of YAP/TAZ. We compared SK-N-MC cells transfected with a control non-targeting siRNA with cells transfected with a mix of siRNA targeting both YAP and TAZ.

GSE73092 High-throughput RNAi cell viability screen to identify selective targets for EWS-FLI1 positive Ewing sarcoma
Contributors : Kristiina Iljin ; Tao HeSeries Type : Expression profiling by high throughput sequencingOrganism : Homo sapiensWe have performed a high-throughput RNA interference screen to identify targets inhibiting EWS-FLI1 driven cell proliferation in Ewing sarcoma cells. EWS-FLI1 expressing A673 Ewing sarcoma cells were screened both in presence and absence of EWS-FLI1 shRNA induction with druggable siRNA library. Leucine rich repeats and WD repeat Domain containing 1 (LRWD1) targeting siRNA pool was the strongest anti-proliferative hit identified only in presence of EWS-FLI1. Validation experiments confirmed the anti-...

GSE93677 Identification of a Novel SYK/c-MYC/MALAT1 Signaling Pathway and Its Potential Therapeutic Value in Ewing Sarcoma

Tyrosine kinases (TKs) and associated pathways are continuously activated in many malignancies including EWS; these enzymes provide candidate therapeutic targets. Using two high-throughput screens with EWS cell lines (a siRNA library and...

GSE73422 Expression data from human osteosarcoma cells treated with CDK11 siRNA
Contributors : Yong Feng ; Zhenfeng DuanSeries Type : Expression profiling by arrayOrganism : Homo sapiensThe U-2OS and KHOS osteosarcoma cell lines where seeded onto the 100mm cell culture plate, then treated with CDK11 siRNA (40nM) or nonspecificsiRNA (40nM). Change with regular medium 24 hour later. Total RNA was collected from these cells using TRIzol® Reagent (GIBCO Grand Island, NY) according to the manufacturer’s instructions. To account for and eliminate biologic noise, RNA was isolated from three distinct flasks of each cell line and pooled together. RNA quality was determined via ethidium bromide staining foll...

GSE57203 Syngergistic Effect of JQ1 and Rapamycin for Treatment of Human Osteosarcoma
In conclusion, our findings suggest that JQ1, in combination with rapamycin, is an effective chemotherapeutic option for OS treatment. We also show that inhibition of RUNX2 expression by JQ1 partly explains antiproliferative activity of JQ1 in OS cells.
