Cold atmospheric plasma mediated cell membrane permeation and gene delivery-empirical interventions and pertinence
Adv Colloid Interface Sci. 2023 Aug 28;320:102989. doi: 10.1016/j.cis.2023.102989. Online ahead of print.ABSTRACTDelivery of genetic material to cells is an integral tool to analyze and reveal the genetic interventions in normal cellular processes and differentiation, disease development and for gene therapy. It has profound applications in pharmaceutical, agricultural, environmental and biotechnological sectors. The major methods relied for gene delivery or transfection requires either viral vectors or xenogenic carrier molecules, which renders probabilistic carcinogenic, immunogenic and toxic effects. A newly evolved phy...

Stimuli-Responsive Hydrogel Microcapsules Harnessing the COVID-19 Immune Response for Cancer Therapeutics
We present the development of stimuli-responsive microcapsules, loaded with a viral immunogenetic agent, harnessing the immune response against the Coronavirus Disease 2019, COVID-19, to selectively attack liver cancer cells (hepatoma) or recognize breast cancer or hepatoma, by expression of green fluorescence protein, GFP. The pH-responsive microcapsules, modified with DNA-tetrahedra nanostructures, increased hepatoma permeation by 50%. Incorporation of a GFP-encoding lentivirus vector inside the tumor-targeting pH-stimulated miRNA-triggered and Alpha-fetoprotein-dictated microcapsules enables the demonstration of neoplas...

Recent advances in drug delivery approaches for rheumatoid arthritis
Curr Med Chem. 2023 Aug 15. doi: 10.2174/0929867331666230815112818. Online ahead of print.ABSTRACTMorbidity, disability, and healthcare expenses associated with rheumatoid arthritis (RA) impose a considerable health and economical burden on both patients and healthcare systems. This review aimed to examine the pathophysiological aspects of RA that may help design different types of drugs and drug delivery systems. These include monoclonal antibodies, immunoglobulins, tiny chemicals, and transgenes for gene therapy. These novel nanocarrier-based therapies target the underlying biological processes involved in RA while minim...

Emerging 2D pnictogens: a novel multifunctional photonic nanoplatform for cutting-edge precision treatment
Chem Commun (Camb). 2023 Aug 9. doi: 10.1039/d3cc02624h. Online ahead of print.ABSTRACTThe elements of the pnictogen group, known as the 15th (VA) family in the periodic table, including phosphorus (P), arsenic (As), antimony (Sb) and bismuth (Bi), have been widely used by alchemists to treat various diseases since ancient times and hold a pivotal position in the history of medicine, owing to their diverse pharmacological activities. Recently, with the development of modern nanotechnology, pnictogen group elements appear in a more innovative form, namely two-dimensional (2D) pnictogens (i.e. phosphorene, arsenene, and bism...

An intelligent DNA nanomachine for amplified MicroRNA imaging and MicroRNA-Guided efficient gene silencing
Talanta. 2023 Jun 13;265:124820. doi: 10.1016/j.talanta.2023.124820. Online ahead of print.ABSTRACTThe DNA nanomachines as excellent synthetic biological tools have been widely used for the sensitive detection of intracellular microRNA (miRNA) and DNAzyme-involved gene silencing. However, intelligent DNA nanomachines which have the ability to sense intracellular specific biomolecules and respond to external information in complex environments still remain challenging. Herein, we develop a miRNA-responsive DNAzyme cascaded catalytic (MDCC) nanomachine to perform multilayer cascade reactions, enabling the amplified intracell...

Synthesis of Polyethyleneimines from the Manganese Catalyzed Coupling of Ethylene Glycol and Ethylenediamine
We report here a novel method for the synthesis of branched polyethyleneimines from ethylene glycol and ethylenediamine feedstock which are much safer, environmentally benign, commercially available and potentially renewable feedstock. The polymerisation reaction is catalysed by a complex of an earth-abundant metal, manganese and liberates H2O as the only by-product. Our mechanistic studies using a combination of DFT computation and experiment suggest that the reaction proceeds by the formation and subsequent hydrogenation of imine intermediates.PMID:37195140 | DOI:10.1002/anie.202306655

Dynamic Hydrogen-Bonded Zinc Adeninate Framework (ZAF) for Immobilization of Catalytic DNA
Angew Chem Int Ed Engl. 2023 Apr 19:e202302840. doi: 10.1002/anie.202302840. Online ahead of print.ABSTRACTEffective immobilization and delivery of genetic materials is at the forefront of biological and medical research directed toward tackling scientific challenges such as gene therapy, and cancer treatment. Here we present a biologically inspired hydrogen bonded zinc adeninate framework (ZAF) consisting of zinc adeninate macrocycles that self-assemble into a 3D framework via adenine-adenine interactions. ZAF can efficiently immobilize DNAzyme with full protection against enzyme degradation and physiological conditions u...
