A sting in the tail-are antibodies against the C-terminus of Plasmodium falciparum circumsporozoite protein protective?
EMBO Mol Med. 2023 Apr 21:e17556. doi: 10.15252/emmm.202317556. Online ahead of print.ABSTRACTMalaria remains a huge burden on global public health. Annually there are more than 200 million cases with > 600,000 deaths worldwide, the vast majority of which occur within Sub-Saharan Africa (WHO; World Malaria Report, 2021). Malaria disease is the consequence of infection by a protozoan parasite from the genus Plasmodium with most morbidity and mortality caused by P. falciparum. With rates of infection plateauing and rebounding in some areas (in particular, as a result of the disruption caused by the COVID-19 pandemic), the...

Molecular and functional properties of human Plasmodium falciparum CSP C-terminus antibodies
EMBO Mol Med. 2023 Apr 21:e17454. doi: 10.15252/emmm.202317454. Online ahead of print.ABSTRACTHuman monoclonal antibodies (mAbs) against the central repeat and junction domain of Plasmodium falciparum circumsporozoite protein (PfCSP) have been studied extensively to guide malaria vaccine design compared to antibodies against the PfCSP C terminus. Here, we describe the molecular characteristics and protective potential of 73 germline and mutated human mAbs against the highly immunogenic PfCSP C-terminal domain. Two mAbs recognized linear epitopes in the C-terminal linker with sequence similarity to repeat and junction motif...

IFN γ, TNFα polymorphisms and IFNγ serum levels are associated with the clearance of drug-resistant P. falciparum in Malian children
This study investigates host regulatory cytokines and immunogenomic factors associated with the clearance of Plasmodium falciparum carrying a chloroquine resistance genotype. Biological samples from participants of previous drug efficacy trials conducted in two Malian localities were retrieved. The P. falciparum chloroquine resistance transporter (Pfcrt) gene was genotyped using parasite DNA. Children carrying parasites with the mutant allele (Pfcrt-76T) were classified based on their ability to clear their parasites. The levels of the different cytokines were measured in serum. The polymorphisms of specific human genes in...

Genetically modified malaria parasites as vaccine candidates
Trends Mol Med. 2022 Nov 17:S1471-4914(22)00295-7. doi: 10.1016/j.molmed.2022.11.005. Online ahead of print.ABSTRACTInduction of sterile immunity against sporozoite and liver stages of malaria is a long-standing aim in vaccine development. Genetically engineered, attenuated sporozoites were systematically evaluated in animal models. Murphy et al. present the first, promising clinical trial of early arresting parasites, PfGAP3KO, confirming safety and demonstrating efficacy against homologous challenge.PMID:36404198 | DOI:10.1016/j.molmed.2022.11.005

Expression of Large Full-Length PfEMP1 Proteins in HEK293 Cells
Methods Mol Biol. 2022;2470:283-298. doi: 10.1007/978-1-0716-2189-9_21.ABSTRACTPlasmodium falciparum erythrocyte membrane protein 1 (PfEMP1) is a family of proteins expressed on the surface of red blood cells infected by Plasmodium falciparum. PfEMP1 proteins play a vital role in parasite virulence, and thus are important vaccine candidates to prevent severe disease. VAR2CSA is one specific PfEMP1 essential for pregnancy malaria pathogenesis, and the primary target in pregnancy malaria vaccine development. However, similar to other PfEMP1 proteins, expression of recombinant full-length VAR2CSA is difficult due to its large...

Production of PfEMP1-Specific Human Monoclonal Antibodies from Naturally Immune Individuals
Methods Mol Biol. 2022;2470:407-421. doi: 10.1007/978-1-0716-2189-9_30.ABSTRACTPlasmodium falciparum parasites express variable surface antigens on the infected erythrocyte surface allowing adhesion to human host receptors on the blood and endothelial cells, which can result in immune evasion. One of the most studied and key antigens in adhesion is the highly polymorphic PfEMP1. However, despite the vast variation in the PfEMP1 antigens, they are the main targets of naturally acquired immunity and are therefore promising candidates for malaria vaccine development. Generating PfEMP1-specific human monoclonal antibodies from...
