This study demonstrates just how vital the thymus is to maintaining adult health." « Back to Top Does Amyloid-β Aggregation Cause Broad Disruption of Proteostasis? https://www.fightaging.org/archives/2023/08/does-amyloid-%ce%b2-aggregation-cause-broad-disruption-of-proteostasis/ Researchers here speculate on the ability of insoluble amyloid-β aggregates to be broadly disruptive of the solubility of many other proteins, and thus disruptive to cell and tissue function. Is this important in aging? The evidence here shows the existence of the mechanism in a lower species, but that doesn't n...

N-terminal alanine-rich (NTAR) sequences drive precise start codon selection resulting in elevated translation of multiple proteins including ERK1/2
We report the discovery of N-terminal alanine-rich sequences, which we term NTARs, that act in concert with their native 5'-untranslated regions to promote selection of the proper start codon. NTARs also facilitate efficient translation initiation while limiting the production of non-functional polypeptides through leaky scanning. We first identified NTARs in the ERK1/2 kinases, which are among the most important signaling molecules in mammals. Analysis of the human proteome reveals that hundreds of proteins possess NTARs, with housekeeping proteins showing a particularly high prevalence. Our data indicate that several of ...

This study explored the association between different cooking fuel types and the risk of cancer and all-cause mortality among seniors constructing Cox regression models. Data were obtained by linking waves of 6, 7, and 8 of the Chinese Longitudinal Healthy Longevity Survey, which included a total of 7,269 participants who were 65 years old and over. Cooking fuels were categorized as either biomass, fossil, or clean fuels. And the effects of switching cooking fuels on death risk were also investigated using Cox regression models. The results indicate that, compared with the users of clean fuels, individuals using bio...

Flutamide With or Without PROSTVAC in Non-metastatic Castration Resistant (M0) Prostate Cancer
CONCLUSION: The combination of flutamide+PROSTVAC did not improve outcomes in men with nmCRPC compared with flutamide alone. (ClinicalTrials.gov Identifier: NCT00450463).PMID:37134294 | DOI:10.1093/oncolo/oyad058

A Phase I/II trial of HER-2/neu vaccine primed autologous T-cell infusions in patients with treatment refractory HER-2/neu overexpressing breast cancer
CONCLUSIONS: Adoptive transfer of HER2 vaccine primed T-cells was feasible, associated with minimal toxicity, and resulted in an increased overall survival in responding patients.PMID:37093223 | DOI:10.1158/1078-0432.CCR-22-3578

In this study, researchers show that mice lacking a functional ATF4 gene show little to no loss of grip strength and treadmill performance into late life; it is quite an impressive effect size. Assessments of muscle biochemistry do show age-related declines, but to a lesser degree than the controls. How ATF4 knockout functions to produce this outcome is an interesting question. The researchers point out a range of possible downstream and upstream targets that have been implicated in the regulation of muscle growth, but it will clearly require further work to identify the important mechanisms involved. Aging slowly...

Cervical human papillomavirus prevalence according to socioeconomic and demographic characteristics in a large Danish screening population
CONCLUSIONS: We found slightly higher hrHPV prevalences in women with basic education, low income and unemployment. The differences largely disappeared when taking into account marital status as a potential proxy for sexual behaviour. Our findings support a need for targeted information on safe sexual practices and promoting socioeconomic equality in HPV vaccination and cervical cancer screening participation.PMID:37078420 | DOI:10.1177/14034948231168297
