Molecules, Vol. 26, Pages 5001: The Nrf2 Pathway in Ischemic Stroke: A Review
ano Saso Ischemic stroke, characterized by the sudden loss of blood flow in specific area(s) of the brain, is the leading cause of permanent disability and is among the leading causes of death worldwide. The only approved pharmacological treatment for acute ischemic stroke (intravenous thrombolysis with recombinant tissue plasminogen activator) has significant clinical limitations and does not consider the complex set of events taking place after the onset of ischemic stroke (ischemic cascade), which is characterized by significant pro-oxidative events. The transcription factor Nuclear factor erythroid 2-related factor...

Thrombus inhibition and neuroprotection for ischemic stroke treatment through platelet regulation and ROS scavenging
This study may provide a promising therapeutic routine for regulating the microenvironment of ischemic stroke through a combined function of ROS scavenging, microthrombus inhibition, and BBB penetration.PMID:36220787 | DOI:10.1002/cmdc.202200317

Discovery of novel danshensu derivatives bearing pyrazolone moiety as potential anti-ischemic stroke agents with antioxidant activity
In this study, a series of novel danshensu derivatives bearing pyrazolone moieties with dual antioxidant effects were synthesized for the treatment of ischemic stroke. Most compounds exhibited considerable DPPH free radical scavenging ability and neuroprotective activity against H2O2-induced oxidative injury in PC12 neuronal cells, without cytotoxicity. Among these target compounds, Del03 displayed the strongest dose-dependent neuroprotective activity in vitro, directly downregulated intracellular ROS levels, and improved the oxidative stress parameters MDA, SOD, and LDH. Del03 also promoted Nrf2 translocation to the nucle...

Design, synthesis and biological evaluation of indoline derivatives as multifunctional agents for the treatment of ischemic stroke
AbstractIn this work, a series of indoline derivatives as multifunctional neuroprotective agents for battling ischemic stroke were designed, synthesized, and biologically evaluated. In antioxidant assay, all compounds showed significant protective effects against H2O2-induced death of RAW 264.7 cells. In oxygen glucose deprivation/reperfusion (OGD/R)-induced neuronal damage, some compounds significantly elevated the cell survival rate. Among them,7i,7j and7r exerted comparable neuroprotective effects to ifenprodil, and exhibited binding affinity toN-methyl-D-aspartic acid receptors 2B (NMDA-GluN2B). At the concentrations o...

Molecules, Vol. 23, Pages 2401: Neuroprotective Effects of Radix Scrophulariae on Cerebral Ischemia and Reperfusion Injury via MAPK Pathways
Xiaobo Sun Ischemic stroke is a clinically common cerebrovascular disease whose main risks include necrosis, apoptosis and cerebral infarction, all caused by cerebral ischemia and reperfusion (I/R). Ischemia and reperfusion-induced injury of or apoptosis inhibition in human brain tissue may exert an irreplaceable protective effect on ischemic nerves. This process has particular significance for the treatment of stroke patients. However, the development of neuroprotective drugs remains challenging. Radix Scrophulariae, traditionally considered a valuable medicine, has been discovered to have neuroprotective effects. ...

Protective effects of hederagenic acid on PC12 cells against the OGD/R-induced apoptosis via activating Nrf2/ARE signaling pathway
AbstractCerebral ischemic stroke is a severe cause of disability and death among people worldwide. The occlusion of cerebral blood flow has resulted in the infarction due to the oxygen and glucose deprivation. Though timely reperfusion can reduce the infarct size, the oxidative stress triggered in ischemia and reperfusion will induce apoptosis of neurons in penumbra. Nuclear factor erythroid 2-related factor 2 (Nrf2) is a transcription factor regulating the expression of antioxidant enzymes through binding to antioxidant response elements (ARE). The activation of Nrf2/ARE pathway can protect neurons against oxidative stres...
