This study aimed to investigate the role of the long non-coding RNA (lncRNA) LINC00342-207 (LINC00342) in the development and progression of primary hepatocellular carcinoma (HCC). Forty-two surgically resected HCC tissues and corresponding paracancerous tissues were collected from October 2019 to December 2020 and examined for lncRNA LINC00342, microRNA (miR)-19a-3p, miR-545-5p, miR-203a-3p, cell cycle protein D1 (CyclinD1/CCND1), murine double minute 2 (MDM2), and fibroblast growth factor 2 (FGF2) expression. The disease-free survival and overall survival of patients with HCC were followed up. HCC cell lines and the norm...

GSE209814 Gene expression profiles in gastric cancer cells with TM4SF1-AS1 knockdown
Contributors : Hiromu Suzuki ; Hiroshi KitajimaSeries Type : Expression profiling by arrayOrganism : Homo sapiensTo clarify the function of a long noncoding RNA TM4SF1-AS1 in gastric cancer (GC), we carried out a gene expression microarray analysis using GC cell lines HSC45 transfected with a siRNA targeting TM4SF1-AS1 or a control siRNA. Gene ontology analysis revealed that genes associated with interferon signaling and immune response were significantly enriched among downregulated genes.

GSE181249 Transcriptional effects of siRNA-mediated AhR knockdown on TLR7 activation of M-CSF-derived macrophages
Contributors : Miriam Sim ón Fuentes ; Ángeles Domínguez Soto ; Miguel A Vega ; Angel L CorbíSeries Type : Expression profiling by high throughput sequencingOrganism : Homo sapiensAnalysis of the role of AhR on the transcriptional signature of TLR7-activated human M-CSF-dependent monocyte-derived macrophages. Methods: Human Peripheral Blood Mononuclear Cells (PBMC) were isolated from buffy coats from donors over a Lymphoprep gradient according to standard procedures. Monocytes were purified from PBMC by magnetic cell sorting using anti-CD14 microbeads (>95% CD14+ cells). Monocytes from three independent donors (0.5 x ...

GSE223836 Characterization of serotonin-5-HTR1E signaling pathways and its role in cell survival
Contributors : Vinay K Sharma ; Kiersten Campbell ; Xuyu Yang ; Ryan Dale ; Y.P LohSeries Type : Expression profiling by high throughput sequencingOrganism : Homo sapiens5-Hydroxy tryptamine receptor 1E (5-HTR1E) is reported to activate cAMP and ERK pathways via its ligands and binding partners, but the detailed mechanism underlying the serotonin induced 5-HTR1E signaling is still not known. In the present study, we determined the cellular regulators of ERK and cAMP signaling pathways in response to serotonin induced 5-HTR1E activation in 5-HTR1E overexpressing HEK293 cells. We found that Pertussis Toxin (PTX) treatment co...

Effects and regulatory pathway of proopinmelanocortin on feeding habit domestication in mandarin fish
Gene. 2023 Jun 17:147581. doi: 10.1016/j.gene.2023.147581. Online ahead of print.ABSTRACTProopiomelanocortin (POMC) is a hormone precursor, and has been reported to participate in domestication. However, its effects on feeding habit domestication in fish are poorly understood. Mandarin fish (Siniperca chuatsi) feeds solely on live prey fish since first-feeding. In the present study, the high expression of pomc in mandarin fish, both the pomc siRNA and MC4R inhibitor treatments increased the success rate of domestication from live prey fish to dead prey fish and food intake of dead prey fish, suggesting the role of pomc on ...

GSE234780 Knockdown TIMM13 in A549 lung cancer cell line
Contributors : Zhaozhao Zhao ; Ting NiSeries Type : Expression profiling by high throughput sequencingOrganism : Homo sapiensWe depleted TIMM13 using short interfering RNA (siRNA) in A549 lung cancer cells, and found that silencing of TIMM13 could significantly inhibit A549 cell proliferation. Thus, we conducted RNA-seq in the A549 cells transduced with either nontargeting siRNA or two distinct TIMM13-specific siRNAs. We observed widespread gene expression change in A549 cells upon TIMM13 knockdown.

Genes, Vol. 14, Pages 1224: miR-497 Regulates LATS1 through the PPARG Pathway to Participate in Fatty Acid Synthesis in Bovine Mammary Epithelial Cells
In this study, microRNA sequencing was performed on mammary gland tissues collected from cows during early, peak, and late lactation to determine miRNA expression profiles. Differentially expressed miRNA (miR-497) was selected for functional studies of fatty acid substitution. Simulants of miR-497 impaired fat metabolism [triacylglycerol (TAG) and cholesterol], whereas knockdown of miR-497 promoted fat metabolism in bovine mammary epithelial cells (BMECs) in vitro. In addition, in vitro experiments on BMECs showed that miR-497 could down-regulate C16:1, C17:1, C18:1, and C20:1 as well as long-chain polyunsaturated fats. Th...
