Epigenetic silence of HOTAIR contributes to the metastasis of pancreatic cancer via targeting miR-138.
Authors: Nie H, Zhang Y, Xing R, Li M, Mou Y Abstract Pancreatic cancer (PC) is one of the most common malignant tumors in digestive tract. The expression of HOX transcriptantisense RNA (HOTAIR) is positively correlated with TNM staging of PC. Our study aims to investigate the effect and the related mechanism of HOTAIR on the invasion and migration ability of PC cells. We found that the expression of HOTAIR was significantly increased in PC tissues and four PC cell lines (PANC-1, SW1990, CAPAN-1 aand JF305) compared with normal tissues and human pancreatic HPDE6-c7 cells. HOTAIR was then silenced by transfecting si...

UCA1 Regulates the Growth and Metastasis of Pancreatic Cancer By Sponging MiR-135a.
This study is aimed to investigate the role of urothelial carcinoma associated 1(UCA1) in the progression of PC. Our results revealed that long non-coding RNAs (lncRNAs)-UCA1 was overexpressed in PC tissues compared with adjacent histologically normal tissues. Down-regulated level of UCA1 was also detected in five human pancreatic cancer cell lines (SW1990,BxPC-3, MiaPaCa-2, PANC-1, CAPAN-1) compared with normal pancreatic duct epithelial HPDE cells. The proliferation of PC cells was inhibited after UCA1 was suppressed by a lentiviral vector. Cell apoptosis rate was largely promoted by down-regulating UCA1. Further resea...

C1qTNF related protein 4 (CTRP4) promotes pancreatic ductal adenocarcinoma cell proliferation and migration via activation of EGFR-PI3K-NF- κB pathway.
In this study, the role of C1qTNF related protein 4 (CTRP4) in the progression of pancreatic ductal adenocarcinoma (PDAC) was explored. We found that, compared with the adjacent non-cancerous tissues and normal pancreatic cells, CTRP4 was markedly upregulated in human PDAC tissues and cell lines. CTRP4 siRNA or pcDNA-CTRP4 expression vectors were transfected into PANC-1 PDAC cells. CTRP4 knockdown sharply reduced IL-8 secretion, causing inactivation of EGFR/PI3K/NF-κB, and suppressing the proliferation and migration of PANC-1 cells. In contrast, CTRP4 overexpression increased IL-8 secretion and EGFR/PI3K/NF-κB activation...

DKK3 Overexpression Increases Malignant Properties of Head and Neck Squamous Cell Carcinoma Cells.
Authors: Katase N, Nishimatsu SI, Yamauchi A, Yamamura M, Terada K, Itadani M, Okada N, Hassan NMM, Nagatsuka H, Ikeda T, Nohno T, Fujita S Abstract DKK3, a member of dickkopf WNT signaling pathway inhibitor family, is believed to be a tumor suppressor, because of itsreduced expression in cancer cells. However, our previous studies have revealed that DKK3 expression is predominantly observed in head and neck, and oral squamous cell carcinoma (HNSCC/OSCC). Interestingly, HNSCC/OSCC patients with DKK3 expression showed high metastasis rate and poorer survival, and siRNA-mediated knockdown of DKK3 in HNSCC-derived can...

Phosphoglycerate mutase 1(PGAM1) promotes pancreatic ductal adenocarcinoma (PDAC) metastasis by acting as a novel downstream target of PI3K/Akt/mTOR pathway.
This study indicates that PGAM1 is closely associated with clinical metastasis and poor prognosis of PDAC. PGAM1 is considered as a potential therapeutic target in PDAC metastasis. PMID: 29386088 [PubMed - as supplied by publisher]
