The Mutational and Transcriptional Landscapes of Speckle-Type POZ Protein (SPOP) and Androgen Receptor (AR) in a Single-Center pT3 Prostatectomy Cohort
J Environ Pathol Toxicol Oncol. 2024;43(1):15-29. doi: 10.1615/JEnvironPatholToxicolOncol.2023048095.ABSTRACTProstate cancer (PCa) is a heterogeneous disease both clinically and genetically. According to The Cancer Genome Atlas (TCGA), the speckle‑type POZ protein (SPOP) mutant form is one of the significant core subtypes of PCa. However, the prognostic value of SPOP variations remains unknown. As a critical PCa driver and an SPOP-targeted protein, androgen receptor (AR) also plays a role in PCa initiation and progression. Thus, we aimed to analyze the mutational status of SPOP and AR with their transcriptional levels in...

Whole-Genome Comparative Copy Number Alteration Profiling between Malignant Pleural Mesothelioma and Asbestos-Induced Chronic Pleuritis
In this study, we scanned the genomes of individuals MPM and asbestos-induced chronic pleuritis (AICP) to compare and determine copy number alterations (CNAs) between two asbestos-related diseases. We used high-resolution SNP arrays to compare CNA profiles between MPM (n = 55) and AICP (n = 18). DNAs extracted from pleural tissues in both groups. SNP array analysis revealed common losses at 1p, 3p, 6q, 9p, 13q, 14q, 15q, 16q, 22q and frequent gains at chromosomes 1, 3, 5, 7, 8, and 6p, 12q, 15q, 17p, 20q in MPMs (frequencies max 67%-min 30%; these alterations were not detected in AICPs. Besides detecting well-known MPM-ass...

Frequency of Healthy Control Genotype of VDR Gene Polymorphisms in the Saudi Population of the Ha'il Region: A Comparative Study with Worldwide Population
In this study, efforts were made to ascertain the frequency of VDR polymorphisms in the Saudi population of Ha'il region, and then comparison was made for VDR polymorphism rates with other populations of the world. Allele and genotype frequencies of VDR TaqI, ApaI, BsmI and FokI gene was determined. The frequency distribution for the variant allele of VDR TaqI, ApaI, BsmI and FokI was found to be 70, 33, 50 and 25%, respectively. A significant frequency distribution was found for VDR-TaqI, ApaI and FokI variants in comparison with other populations of the world. Whereas, almost all of the studies dealing with VDR-FokI fail...

Can the Toxic Heavy Metals Be Beneficial at Trace Levels? Understanding Their Outranged Biological Functions
This article aims to collate such investigations conducted during the last two decades with trace levels of cadmium, mercury and arsenic toxicity against animal systems. There are a total of 13 pre-clinical works demonstrating the protective or beneficial effect of mercury, cadmium and arsenic at trace levels. Such literature reports with lead could not be found. From the outcome of the current literature investigation it is evident that, trace levels of toxic heavy metals namely arsenic, cadmium and mercury possess remarkable protective and beneficial effects chiefly on growth, developmental and reproductive parameters ag...

Transcription Factor E2F1 Enhances Hepatocellular Carcinoma Cell Proliferation and Stemness by Activating GINS1
This study aims at investigating the mechanism by which GINS1 affects HCC cell proliferation and stemness. We performed bioinformatics analysis for determining GINS1 expression in HCC tissues, as well as the HCC patients' survival rate with different expression levels of GINS1. E2F transcription factor 1 (E2F1) was predicted as the upstream transcription factor of GINS1, and the binding relation between the two was verified by chromatin immunoprecipitation and dual-luciferase reporter assays. Quantitative real-time polymerase chain reaction was adopted to evaluate the expression of GINS1 and E2F1. The protein expression le...

Chemotherapeutic Drugs Endow Gastric Cancer Mesenchymal Stem Cells with Stronger Tumor-Promoting Ability
In this study, flow cytometry, CCK8 assay, migration assay, colony formation assay, and western blot were conducted. We also analyzed GC patients from the cancer genome atlas (TCGA). Our results showed that GC-MSCs were resistant to 5-FU and Taxol at the IC50 concentration for GC cells, and 5-FU could promote the migration of GC-MSCs at low doses. Furthermore, the conditioned medium of GC-MSCs pretreated with chemotherapeutic drugs was more effective in promoting the proliferation, migration, and stemness of GC cell lines than the conditioned medium of GC-MSCs without chemotherapeutic drugs treatment. These effects were de...
