CONCLUSION: The serum level of S100B was related to the presence of vitiligo, but its level did not show any relation to the disease activity using either VASI and VIDA scores or various disease parameters. PMID: 32951206 [PubMed - as supplied by publisher]

Immune checkpoint blockade therapy can induce immune-related toxicity, but cutaneous lymphoma development has not been reported. A 56-year-old woman presented with two well-demarcated erythematous macules on the right sole and vitiligo on her extremities. Her facial melanoma had been treated with combination therapy (ipilimumab and pembrolizumab), followed by pembrolizumab monotherapy, a year prior. Microscopy revealed small-to-medium-sized lymphocytes permeating along with the basal epidermal layer. These were immuno-positive for CD2, CD3, and CD5, and showed complete CD7 loss; CD30, TCR-beta F1, and PD-1 were also detect...

Abstract It is technically impossible to ascertain cumulative UVR exposure with the Korean National Health Insurance claims database due to the lack of information on each treatment dose. As an alternative, we stratified our patients based on the number of phototherapy sessions (no phototherapy,

Abstract PURPOSE: Few studies have investigated the relationship between vitiligo and risks of various types of cancers, especially those other than skin cancer. Conventional observational studies are susceptible to potential confounders and inverse causation. With a Mendelian randomization approach, we were able to evaluate the causality between vitiligo and different cancer risks. METHODS: 37 vitiligo-related single-nucleotide polymorphisms identified by the published genome-wide association studies were used as instrumental variables in our study. Summary data of individual-level genetic information were o...

In metastatic melanoma, both tumor neoantigen load and development of vitiligo have been associated with favorable response to immunotherapy. We hypothesize that in the context of immune checkpoint blockade (ICB), neoantigens facilitate epitope spreading and immune targeting of tumor-lineage self-antigens. We also aim to harness this process with a hapten-based vaccine to improve ICB response. Previously, our lab demonstrated in a murine model of melanoma that tumors with high neoantigen load respond significantly better to ICB than syngeneic tumors with low neoantigen load.

Vitiligo is an autoimmune disease resulting in death of melanocytes, causing disfiguring loss of skin pigmentation. It is known that numerous differentially expressed genes (DEGs) are present in vitiligo lesional skin. However, it is unknown if vitiligo DEGs are correlated with meaningful clinical characteristics, such as disease severity, duration and therapeutic response. The purpose of this study is to evaluate the clinical correlates of vitiligo DEGs. Thirty seven vitiligo patients were recruited for this study, each consented to skin biopsies from lesional as well as non-lesional skin.

Cytoarchitectonic and functional interactions among epidermal and dermal compartments are known to regulate melanocyte behaviour and survival. The presence of a deregulated microenvironment able to affect melanocyte homeostasis has been described in both hyper- and hypo-pigmentary disorders. In vitiligo, several data indicate a structural and functional impairment not only of melanocytes, but also of the other cutaneous cells, which appears broadly extended, even in apparently normal skin. In line with these remarks, our group has recently reported some modifications in non lesional fibroblasts, which show a myofibroblast ...

Vitiligo is a chronic, inflammatory skin disease characterized by increased interferon-gamma signaling through Janus kinase (JAK) 1 and JAK2 and subsequent activation of CD8+ T cells, which target melanocytes resulting in areas of depigmentation. Ruxolitinib cream, a JAK1/JAK2 inhibitor, is under investigation for vitiligo treatment in a 52-week, randomized, double-blind, phase 2 study (NCT03099304). Significantly more patients treated with ruxolitinib cream vs vehicle achieved ≥50% improvement in facial Vitiligo Area Scoring Index (FVASI50) at Week 24 (primary endpoint); at Week 52, patients treated with ruxolitinib cr...

Authors: Maria Alexandra Stanescu A, Totan A, Mircescu D, Diaconescu S, Gabriel Bratu O, Fekete L, László Fekete G, Boda D, Cristina Diaconu C Abstract Many diseases that affect physical health can also affect mental health. Many of these diseases are studied in terms of quality of life, depression and suicidal behavior. Dermatological disorders, although having a strong impact on the individual's life, are less studied with regard to suicidal behavior. There is a need to approach several dermatological diseases from the perspective of suicidal behavior in order to intervene early with specific treatm...

In this study, we conducted a retrospective study of advanced melanoma patients who received PD-1 inhibitor therapy in China between August 2014 and March 2018. A total of 93 patients treated with PD-1 inhibitors including pembrolizumab and nivolumab were enrolled. The most frequent irAEs were pruritus, rash, vitiligo, and fatigue. The median time to onset of irAEs was 6.1 weeks. The overall response rate (ORR) and disease control rate (DCR) were higher in patients with irAEs than those without irAEs (P = 0.004 and P = 0.003, respectively), and better in patients who experienced three or more irAEs than those with none (P
