Expression, purification and crystallization of an SLC16 monocarboxylate transporter family homologue specific for l-lactate

BosshartAbstractl-lactate plays an important role as metabolite and signaling molecule in eukaryotes and bacteria. Monocarboxylate transporters (MCTs) of the SLC16 solute carrier family are responsible for the transport of l-lactate across eukaryotic and bacterial cell membranes. Here we report an efficient protocol for the expression and purification of an SLC16 family homologue in milligram amounts. The purified protein is stable and can thus be used for biochemical and structural studies as shown by successful crystallization.
