Performance evaluation of a line blot assay system for detection of anti ‐PM‐Scl antibody in Japanese patients with systemic sclerosis

ConclusionDetection of anti ‐PM‐Scl Ab assessed with a commercial LB assay requires extreme caution since it may yield false‐positive data.

Systemic sclerosis (SSc) is a connective tissue disorder characterized by the development of fibrosis in the skin and internal organs, as well as microvascular dysfunction [1,2]. Vasculopathy occurs at an early step of SSc. Almost all patients present Raynaud's phenomenon as an initial symptom, and frequently show other manifestations of vascular diseases, including ischemic digital ulcers, pulmonary arterial hypertension, and renal arterial involvement associated with malignant hypertension and renal failure.

AbstractRaynaud ’s phenomenon (RP) is used to describe a symptom complex caused by digital vascular compromise. RP is a clinical diagnosis. The typically episodic nature of RP has resulted in a reliance upon patient self-report for diagnosis. The term ‘primary RP’ is generally applied when no underlying patho logy can be demonstrated. Whilst ‘primary RP’ is currently considered a distinct disorder, there is evidence that the term may comprise several entities that include a functional vasospastic disorder, a physiologically appropriate thermoregulatory response, subclinical atherosclerosis and...

Both Systemic Lupus Erythematosus (SLE) and Systemic Sclerosis (SSc) are autoimmune diseases sharing similar genetic backgrounds. Genome-wide association studies (GWASs) have constantly disclosed numerous genetic variants conferring to both disease risks at 7q32.1, but the functional mechanisms underlying them are still largely unknown. Through a series of bioinformatics and functional analyses, we prioritized a potential independent functional SNP (rs13239597) within TNPO3 promoter region, residing in a putative enhancer element and validated that IRF5 is the distal target gene ( ∼118kb) of rs13239597, which is a key ...

Abstract OBJECTIVE: Pulmonary arterial hypertension (PAH) is one of the leading causes of death in systemic sclerosis (SSc). Current screening algorithms are hampered by low positive predictive values. Outcome measures that could futurely add to performance characteristics would be very welcome. Against this background, we aim to evaluate the role of nailfold videocapillaroscopy (NVC) using standardized definitions, in SSc related PAH (SSc-PAH). METHODS: A systematic review to identify original research papers documenting an association between NVC and right heart catheterisation defined SSc-PAH was performed...
