Histone deacetylase inhibitors (HDACis) prevent chromatin condensation and affect gene transcription. HDACis increase the activity of the P53 tumor suppressor but block T- and B-cell transcription factors, resulting in cell cycle arrest and apoptosis of leukocytes responding to antigen. HDACis work synergistically with alkylating agents because an open chromatin structure allows alkylating agents better access to DNA. Dooley and colleagues tested EDO-S101, a dual HDACi and alkylating agent, in 2 models of myeloperoxidase (MPO) vasculitis.
