Synthesis, characterization and biological applications of substituted pyrazolone core based platinum(II) organometallic compounds

Patel New series of organometallic platinum(II) compounds (I-VI) containing the 2-phenyl-5-methyl pyrazole-3-one derivatives ligands (L1-L6) were synthesized and characterized by spectroscopic and physicochemical techniques. The synthesized compounds were screen for their in vitro antibacterial activity against two Gram(+ve) and three Gram(−ve) bacterial species. In vitro cytotoxicity against brine shrimp lethality bioassay and in vitro cellular level cytotoxicity against S. pombe cells were carried out for the synthesized compounds. Complexes (I-VI) exhibit excellent cytotoxicity as compared to ligands (L1-L6). DNA interaction and DNA cleavage study of the synthesized ligands and complexes were carried out using absorption titration spectroscopy, fluorescence quenching analysis, viscosity measurements, molecular docking study and agarose gel electrophoresis. The interactions of compounds towards herring sperm DNA (HS-DNA) was examined. The binding constant (K b), Stern-Volmer quenching constant (K sv) and associative binding constant (K a) of the synthesized ligands and complexes were observed in the range of 0.138–5.580 × 105 M−1, 1.1 × 102 to 7.0 × 103 M−1 and 1.871 × 103 to 1.711 × 105 M−1, respectively.
