Sample Preparation and Phosphopeptide Enrichment for Plant Phosphoproteomics via Label-Free Mass Spectrometry

Methods Mol Biol. 2024;2787:293-303. doi: 10.1007/978-1-0716-3778-4_20.ABSTRACTPhosphopeptide enrichment is the main bottleneck of every phosphorylation study. Therefore, in this chapter, a general workflow tries to overbridge the hurdles of plant sample handling from sample collection to protein extraction, protein solubilization, enzymatic digestion, and enrichment step prior to mass spectrometry. The workflow provides information to perform global proteomics as well as phosphoproteomics enabling the researcher to use the protocol in both fields.PMID:38656498 | DOI:10.1007/978-1-0716-3778-4_20
