GATA3 binds to the Cftr locus in CD4+ T cells

Previously we have shown greater type helper 2 (Th2) cell polarization and effector function in cystic fibrosis transmembrane conductance regulator null (Cftr-/-) compared to Cftr+/+ CD4+ T cells with IL-4 or IL-33 pathway activation (Cook et al. AJRCCM. 2023). Given that GATA3 is the master transcriptional regulator of Th2 function, we hypothesized 1) increased GATA3 expression in Cftr-/- compared to Cftr+/+ CD4+ T cells and 2) binding of GATA3 to the Cftr locus to regulate CFTR expression.
