DL-3-n-butylphthalide improves the endothelium-dependent vasodilation in high-fat diet-fed ApoE < sup > -/- < /sup > mice via suppressing inflammation, endothelial necroptosis and apoptosis
This study aims to explore whether NBP is able to improve endothelium-dependent vasodilation in atherosclerosis and the underlying mechanisms. Male ApoE-/- mice were fed with a high-fat diet (HFD) for 9-16 weeks to establish a model of atherosclerosis. NBP were given to the mice after eating HFD for 6 weeks and atorvastatin served as a positive control. The endothelium-dependent vasodilation, the blood flow velocity, the atherosclerotic lesion area, the serum levels of lipids, inflammatory cytokines and necroptosis-relevant proteins (RIPK1, RIPK3 and MLKL), and the endothelial necroptosis and apoptosis within the aorta wer...

The multiple roles of lysophosphatidic acid in vascular disease and atherosclerosis
Curr Opin Lipidol. 2023 Jul 28. doi: 10.1097/MOL.0000000000000890. Online ahead of print.ABSTRACTPURPOSE OF REVIEW: To explore the multiple roles that lysophosphatidic acid (LPA) plays in vascular disease and atherosclerosis.RECENT FINDINGS: A high-fat high-cholesterol diet decreases antimicrobial activity in the small intestine, which leads to increased levels of bacterial lipopolysaccharide in the mucus of the small intestine and in plasma that increase systemic inflammation, and enhance dyslipidemia and aortic atherosclerosis. Decreasing LPA production in enterocytes reduces the impact of the diet. LPA signaling inhibit...

A cyclic azapeptide ligand of the scavenger receptor CD36/SR-B2 reduces the atherosclerotic lesion progression and enhances plaque stability in apolipoprotein E-deficient mice
Front Pharmacol. 2023 May 30;14:1204905. doi: 10.3389/fphar.2023.1204905. eCollection 2023.ABSTRACTAtherosclerosis is a chronic inflammatory disease of the arterial walls that develops at predisposed sites. As a major risk factor for adverse cardiovascular pathology, atherosclerosis can progress to myocardial infarction and stroke, due to the rupture of unstable atherosclerotic lesions. Macrophage uptake of modified lipoproteins and metabolic dysfunction contributes significantly to the initiation and development of atherosclerotic lesions. The cluster of differentiation 36 receptor [CD36 (SR-B2)] plays a key role in ather...

Endothelial CCRL2 induced by disturbed flow promotes atherosclerosis via chemerin-dependent β2 integrin activation in monocytes
CONCLUSIONS: Our findings indicate that d-flow-induced CCRL2 promotes atherosclerotic plaque formation via a novel CCRL2-chemerin-β2 integrin axis, providing potential targets for the prevention or therapeutic intervention of atherosclerosis.PMID:37279540 | DOI:10.1093/cvr/cvad085

Pathways linking aging and atheroprotection in Mif-deficient atherosclerotic mice
FASEB J. 2023 Mar;37(3):e22752. doi: 10.1096/fj.202200056R.ABSTRACTAtherosclerosis is a chronic inflammatory condition of our arteries and the main underlying pathology of myocardial infarction and stroke. The pathogenesis is age-dependent, but the links between disease progression, age, and atherogenic cytokines and chemokines are incompletely understood. Here, we studied the chemokine-like inflammatory cytokine macrophage migration inhibitory factor (MIF) in atherogenic Apoe-/- mice across different stages of aging and cholesterol-rich high-fat diet (HFD). MIF promotes atherosclerosis by mediating leukocyte recruitment, ...

Mechanism of Tao Hong Decoction in the treatment of atherosclerosis based on network pharmacology and experimental validation
CONCLUSION: THD plays a vital role in treating AS with multiple targets and pathways. Angiogenesis regulation, oxidative stress regulation, and immunity regulation consist of the crucial regulation cores in the mechanism. This study identified essential genes and pathways associated with the prognosis and pathogenesis of AS from new insights, demonstrating a feasible method for researching THD's chemical basis and pharmacology.PMID:36776258 | PMC:PMC9909180 | DOI:10.3389/fcvm.2023.1111475
