Evaluating how cationic lipid affects mRNA-LNP physical properties and biodistribution
Eur J Pharm Biopharm. 2023 Aug 12:S0939-6411(23)00205-9. doi: 10.1016/j.ejpb.2023.08.002. Online ahead of print.ABSTRACTRNA therapeutics represents a powerful strategy for diseases where other approaches have failed, especially given the recent successes of mRNA vaccines against the coronavirus disease 2019 (COVID-19) and small interfering (siRNA) therapeutics. However, further developments are still required to reduce toxicity, improve stability and biodistribution of mRNA-LNPs (lipid nanoparticles). Here, we show a rational combinatorial approach to select the best formulation based on a new cationic lipid molecule (IM21...

Biophysical characterization of siRNA-loaded Lipid Nanoparticles with different PEG content in an aqueous system
This study evaluated the influence of PEGylation and stress conditions on the stability of siRNA-loaded LNPs with different concentrations of PEG (0.5 mol%; 0.5% PEG-LNP and 1.0 mol%; 1.0% PEG-LNP) anchored to their surface. We applied end-over-end agitation, elevated temperature, and repeated freeze and thaw (F/T) cycles as physicochemical stressors of pH and ionic strength. Dynamic light scattering (DLS), flow imaging microscopy (FIM), and ionic-exchange chromatography (IEX) were to determine the degree of aggregation and change in siRNA content. The results indicate that 0.5% PEG-LNP resisted aggregation only at low pH ...

Microfluidic mixing as platform technology for production of chitosan nanoparticles loaded with different macromolecules
Eur J Pharm Biopharm. 2023 May 15:S0939-6411(23)00124-8. doi: 10.1016/j.ejpb.2023.05.010. Online ahead of print.ABSTRACTIn recent decades, biotechnological drugs have emerged as relevant therapeutic tools. However, therapeutic molecules can exert their activity only if properly formulated and delivered into the body. In this regard, nano-sized drug delivery systems have been shown to provide protection, stability, and controlled release of payloads, increasing their therapeutic efficacy. In this work, a microfluidic mixing technique for the preparation of chitosan-based nanoparticles was established with the capability of ...

Multivalent mannose-conjugated siRNA causes robust gene silencing in pancreatic macrophages in vivo
Eur J Pharm Biopharm. 2023 Jan 2:S0939-6411(22)00322-8. doi: 10.1016/j.ejpb.2022.12.017. Online ahead of print.ABSTRACTNucleic acid therapeutics have been utilized for gene regulation, and their recent advancement has led to approval of novel drugs for liver-related disorders. However, systemic extrahepatic delivery remains challenging. Here, we report newly designed mannose-conjugated oligonucleotides for delivering oligonucleotides to macrophages by leveraging the mannose receptor, C-type 1 (MRC1, CD206), which is abundantly expressed in macrophages. We investigated the relationship between cellular uptake and multivalen...

Inhibitory Effect of Aptamer-Carbon Dot Nanomaterial-siRNA Complex on the Metastasis of Hepatocellular Carcinoma Cells by Interfering with FMRP
Eur J Pharm Biopharm. 2022 Mar 29:S0939-6411(22)00061-3. doi: 10.1016/j.ejpb.2022.03.013. Online ahead of print.ABSTRACTUsing small interfering RNA (siRNA) for the specific gene-silencing has been a novel therapeutic method for the treatment of incurable diseases such as malignancies. However, it remains a challenge whether siRNA can be safely and effectively delivered into target cells. Therefore, we synthesized fluorescent carbon dots (CDs) as a gene vector at the siRNA delivery system that induced efficient gene knockdown in vitro while binding aptamer AS1411 to resolve the difficulty in cell targeting. We found that CD...

Mitochondria-Targeted CoQ < sub > 10 < /sub > Loaded PLGA-b-PEG-TPP Nanoparticles: Their Effects on Mitochondrial Functions of COQ8B < sup > -/- < /sup > HK-2 cells
Eur J Pharm Biopharm. 2022 Feb 26:S0939-6411(22)00039-X. doi: 10.1016/j.ejpb.2022.02.018. Online ahead of print.ABSTRACTCoenzyme Q10 (CoQ10) deficiency exhibits signs of multiple organ dysfunctions, particular subtypes present isolated kidney involvement progressing to chronic kidney disease. In these patients, early administration of oral CoQ10 has been shown to decrease proteinuria and to delay development of chronic kidney disease, which suggests that it may have a renoprotective potential in these patients. However, CoQ10 bioavailability in mitochondria is low, therefore its efficacy is limited. We aimed to develop mit...
