Signal detection of adverse events associated with gabapentinoid use for chronic pain
CONCLUSIONS: Our study identified expected and unexpected ADE signals in GABA users. Neurological signals likely related to indications for GABA use. Signals for immunity, mental/behavior, and skin disorders were found in the FDA adverse event reporting system database. Unexpected signals of stroke and cancer require further confirmatory analyses to verify.PMID:37640024 | DOI:10.1002/pds.5685

Tumor Genomic Profile Is Associated With Arterial Thromboembolism Risk in  Patients With Solid Cancer
CONCLUSIONS: In a large genomic tumor-profiling registry of patients with solid cancers, alterations in KRAS and STK11 were associated with an increased risk for ATE independent of cancer type. Further investigation is needed to elucidate the mechanism by which these mutations contribute to ATE in this high-risk population.PMID:37144118 | PMC:PMC10152200 | DOI:10.1016/j.jaccao.2023.01.009

Hypoxanthine is a pharmacodynamic marker of ischemic brain edema modified by glibenclamide
Cell Rep Med. 2022 Jun 9:100654. doi: 10.1016/j.xcrm.2022.100654. Online ahead of print.ABSTRACTBrain edema after a large stroke causes significant morbidity and mortality. Here, we seek to identify pharmacodynamic markers of edema that are modified by intravenous (i.v.) glibenclamide (glyburide; BIIB093) treatment. Using metabolomic profiling of 399 plasma samples from patients enrolled in the phase 2 Glyburide Advantage in Malignant Edema and Stroke (GAMES)-RP trial, 152 analytes are measured using liquid chromatography-tandem mass spectrometry. Associations with midline shift (MLS) and the matrix metalloproteinase-9 (MM...
