Magic shotgun approach to anti-inflammatory pharmacotherapy: Synthesis of novel thienopyrimidine monomers/heterodimer as dual COX-2 and 15-LOX inhibitors endowed with potent antioxidant activity
Eur J Med Chem. 2023 Aug 15;260:115724. doi: 10.1016/j.ejmech.2023.115724. Online ahead of print.ABSTRACTEmerging evidence points to the intertwining framework of inflammation and oxidative stress in various ailments. We speculate on the potential impact of the magic shotgun approach in these ailments as an attempt to mitigate the drawbacks of current NSAIDs. Hence, we rationally designed and synthesized new tetrahydrobenzo[4,5]thieno[2,3-d]pyrimidine monomers/heterodimer as dual selective COX-2/15-LOX inhibitors with potent antioxidant activity. The synthesized compounds were challenged with diverse in vitro biological as...

Diclofenac and eugenol hybrid with enhanced anti-inflammatory activity through activating HO-1 and inhibiting NF- κB pathway in vitro and in vivo
Eur J Med Chem. 2023 Jul 19;259:115669. doi: 10.1016/j.ejmech.2023.115669. Online ahead of print.ABSTRACTA series of diclofenac hybrid molecules were synthesized and evaluated for their NO-inhibitory ability in LPS-induced RAW 264.7 macrophage cells. Among them, compound 1 showed the highest NO-inhibitory ability (approximately 66%) and no significant cytotoxicity. Compound 1 exhibited superior NF-κB-inhibitory ability compared to diclofenac through the activation of Nrf2/HO-1 signaling pathway in RAW 264.7. 20 mg/kg compound 1 resulted in remarkable colitis improvement in dextran sulfate sodium (DSS)-induced mice model b...

Occurrence and removal rate of typical pharmaceuticals and personal care products (PPCPs) in an urban wastewater treatment plant in Beijing, China
Chemosphere. 2023 Jul 24:139644. doi: 10.1016/j.chemosphere.2023.139644. Online ahead of print.ABSTRACTThe occurrence and removal rate of 52 typical pharmaceuticals and personal care products (PPCPs) were investigated in a wastewater treatment plant in Beijing, China. Thirty-three PPCPs were found in the influent, with caffeine (CF, 11387.0 ng L-1) being the most abundant, followed by N,N-diethyl-meta-toluamide (DEET, 9568.4 ng L-1), metoprolol (MTP, 930.2 ng L-1), and diclofenac (DF, 710.3 ng L-1). After treatment processes, the cumulative concentration of PPCPs decreased from 2.54 × 104 ng L-1 to 1.44 × 103 ng L-1, wit...

Effect of salinity on the toxicity of diclofenac, ibuprofen and naproxen toward cyanobacterium Synechocystis salina
In this study, we decided to check how selected non-steroidal anti-inflammatory drugs (NSAID) affect the grow of Synechocystis salina, picocyanobacterium isolated from the Baltic Sea, with salinity as potential modulator of toxicity. S. salina was exposed to diclofenac (DCF), ibuprofen (IBF) and naproxen (NPX) (nominal 100 mg L-1) in BG11 medium and sea salt supplemented BG11 medium (38 PSU) over 96 h in continuous light at 23 °C. No acute toxicity was found in both tested salinity levels. The comparable grow rate in exposed culture compared to control culture over 4 days indicate lack of stress for several generations wh...
