The Impact of Rapid Drug Susceptibility Tests on Gonorrhea Burden and Lifespan of Antibiotic Treatments: A Modeling Study Among Men Who Have Sex With Men in the United States
Am J Epidemiol. 2023 Aug 24:kwad175. doi: 10.1093/aje/kwad175. Online ahead of print.ABSTRACTRapid point-of-care tests that diagnose gonococcal infections and identify susceptibility to antibiotics enable individualized treatment. This could improve patient outcomes and slow the emergence and spread of resistance. However, little is known about the long-term impact of such diagnostics on the burden of gonorrhea and the effective lifespan of antibiotics. We used a mathematical model of gonorrhea transmission among men who have sex with men in the US to project the annual rate of reported gonorrhea cases and the effective li...

United States' regulatory approved pharmacotherapies for nuclear reactor explosions and anthrax-associated bioterrorism
Expert Opin Drug Saf. 2023 Aug 18:1-6. doi: 10.1080/14740338.2023.2245748. Online ahead of print.ABSTRACTINTRODUCTION: Nuclear reactor incidents and bioterrorism outbreaks are concerning public health disasters. Little is known about US Food and Drug Administration (FDA)-approved agents that can mitigate consequences of these events. We review FDA data supporting regulatory approvals of these agents.AREAS COVERED: We reviewed pharmaceutical products approved to treat Hematopoietic Acute Radiation Syndrome (H-ARS) and to treat or prevent pulmonary infections following Bacillus anthracis (anthrax) exposure. Four drugs were a...

Production and certification of BOTS-1: bovine muscle-certified reference material for incurred veterinary drug residues
Anal Bioanal Chem. 2023 Jun 16. doi: 10.1007/s00216-023-04794-5. Online ahead of print.ABSTRACTA freeze-dried bovine muscle-certified reference material (CRM), known as BOTS-1 (DOI: https://doi.org/10.4224/crm.2018.bots-1 ), containing incurred residues of commonly used veterinary drugs was produced and certified for the mass fraction of eight veterinary drug residues. Value assignment was carried out using liquid chromatography tandem mass spectrometry (LC-MS/MS) methods in conjunction with isotope dilution and standard addition approaches involving stable isotope internal standards. Data from the National Research Counci...

Drug-induced liver injury with commonly used antibiotics in the All of Us Research Program
Clin Pharmacol Ther. 2023 May 7. doi: 10.1002/cpt.2930. Online ahead of print.ABSTRACTAntibiotics are a known cause of idiosyncratic drug-induced liver injury (DILI). According to CDC, the five most commonly prescribed antibiotics in the United States are azithromycin, ciprofloxacin, cephalexin, amoxicillin, and amoxicillin-clavulanate. We quantified the frequency of acute DILI for these common antibiotics in the All of Us Research Program, one of the largest Electronic Health Record (EHR)-linked research cohorts in the United States. Retrospective analyses were conducted applying a standardized phenotyping algorithm to de...

Development of Modernized Acinetobacter baumannii Susceptibility Test Interpretive Criteria for Recommended Antimicrobial Agents Using Pharmacometric Approaches
Antimicrob Agents Chemother. 2023 Mar 22:e0145222. doi: 10.1128/aac.01452-22. Online ahead of print.ABSTRACTAcinetobacter baumannii-Acinetobacter calcoaceticus complex (referred to herein as A. baumannii) treatment guidelines contain numerous older antimicrobial agents with susceptibility test interpretive criteria (STIC, also known as susceptibility breakpoints) set using only epidemiological data. We utilized a combination of in vitro surveillance data, preclinical murine thigh and lung infection models, population pharmacokinetics, simulation, and pharmacokinetic/pharmacodynamic (PK/PD) target attainment analyses to eva...

Outpatient antibiogram and predictors of ciprofloxacin and trimethoprim-sulfamethoxazole resistant urinary tract infections
Conclusion: The Infectious Disease Society of America cautions against prescribing an antibiotic if regional resistance exceeds 20%. We constructed an antibiogram and found resistance surpassed this threshold for TMP-SMX and ciprofloxacin and identified factors associated with resistance to these agents. Assessing these characteristics during clinical decision making may improve antibiotic-organism susceptibility concordance in primary care.PMID:36944052 | DOI:10.1370/afm.20.s1.3177
